摘 要:目的探讨伴有kazal域的富含半胱氨酸的逆转诱导蛋白(RECK)与基质金属蛋白酶9(MMP-9)在人骨肉瘤组织中的表达及相关性。方法选取唐山市人民医院2001年1月~2010年12月临床及病理资料完整存档的人骨肉瘤组织标本34例为观察组,另取临床及病理资料完整存档的骨软骨瘤组织标本34例为对照组,采用免疫组织化学法检测RECK和MMP-9,采用定量PCR法检测RECK和MMP-9的mRNA,并分析指标相关性。结果观察组RECK阳性率(58.8%)低于对照组(91.2%),而MMP-9阳性率(64.7%)高于对照组(11.8%),差异有统计学意义(P<0.05)。RECK阳性率、RECK的mRNA表达、MMP-9阳性率、MMP-9的mRNA表达在年龄、性别、病发部位方面比较,差异无统计学意义(P>0.05)。RECK阳性率、RECK的mRNA表达、MMP-9阳性率、MMP-9的mRNA表达在转移、复发以及生存时间方面比较,差异有统计学意义(P<0.05)。RECK与人骨肉瘤组织呈明显负相关(r=-0.561,P=0.000)。MMP-9与人骨肉瘤组织呈明显正相关(r=0.613,P=0.000)。结论 RECK可抑制MMP-9表达,发挥抑癌作用,有望成为未来诊断以及治疗人骨肉瘤的潜在靶标。Objective To explore the expression and correlation analysis of RECK and MMP-9 in human osteosarcoma tissues. Methods 34 c.,ases of human osteosarcoma tissues which had complete clinical and pathological data were selected in Tangshan People's Hospital from January 2001 to December 2010 as observation group. 34 cases of osteuclmmlmma tissues which had complete clinical and pathological data were selected as control group, hnmunohisto- chemieal method was used to detect RECK and MMP-9. Quantitative PCR method was used to detect mRNA of RECK aml M~]P-9. Correlation of indexes were analyzed. Results Positive rate of RECK in observation group (58.8%) was lower than lhat of control group (91.2%), positive rate of MMP-9 in observation group (64.7%) was higher than that of ~:nntrol group (11.8%), with statistical dift^rence (P 〈 0.05). As RECK positive rate, mRNA expression of RECK, M MP-9 positive rate, mRNA expression of MMP-9 compared in gender, age, disease location, differences were not sta- listically significant (P 〉 0.05). As RECK positive rate, mRNA expression of RECK, MMP-9 positive rate, mRNA expression of MMP-9 compared in survival time, recurrence, metastasis, differences were statistically significant (P 〈 0.05). RECK was negatively correlated with human osteosarcoma tissues (r=-0.561, P=0.000). MMP-9 was positively correlated with human osteosarcoma tissues (r=0.613, P=0.000). Conclusion RECK can inhibit the expression of MMP-9, and play a role in inhibiting cancer, which can be expected to potential target for future diagnosis and treatment in human osleusarcnnla.
