摘 要:目的观察体外培养星形胶质细胞(asrocytes,AS)在不同浓度干扰素β(IFN-β)作用下,细胞形态、细胞连接的变化和细胞骨架及细胞弹性相关蛋白表达情况,研究内源性IFN-β对AS的免疫调节作用。方法将体外培养的AS分为对照组和实验组,其中实验组根据IFN-β不同的浓度(102U/mL和103U/mL)及不同的作用时间分为6组。用免疫荧光法鉴定AS,用原子力显微镜观察细胞形态,电镜观察细胞连接,MTT检测IFN-β对细胞生长情况的影响,Western blot观察细胞结构及细胞弹性相关蛋白表达情况。结果随着IFN-β作用时间延长及药物浓度增加,细胞连接增多,细胞骨架及细胞弹性相关蛋白表达也增强。结论在IFN-β作用下AS弹性及韧性增强,细胞突触增加,连接更加紧密,对调节中枢神经系统(central nervous system,CNS)的血脑屏障(blood brain barrier,BBB)的通透性有着重要的意义。Objective To investigate the role of interferon beta (IFN-β) at different times on cell growth state, cell morphology, cell connections associated with protein expression of the astrocytes (AS) cultured in vitro. Methods The cultured AS in vitro was divided into control and experimental groups, and the experimental groups were divided into six groups according to different concentrations of IFN-β (10^2 U/mL and 10^3 U/mL) and different times. AS was identified by immunofluorescenee. The cell morphology was observed by atomic force micro- scope. The cell junction was studied by electron microscopy and the cell growth was observed by MTT. The expression of cytoskeleton-elasticity associated with proteins were analyzed by Western blot. Results There was a obvious increase in the cell connections and the expression of cytoskeleton-elasticity associated protein. Conclusion IFN-β may enhance the flexibility and toughness of AS, and make the connections of AS more closely, which is important in the central nervous system (CNS) to maintain the blood-brain barrier permeability and prevent the CNS from demyelination.
