摘 要:目的研究去甲斑蝥素(norcantharidin,NCTD)作用于体外急性淋巴细胞白血病细胞株Jurkat后survivin及p53 mRNA表达的变化,进一步明确NCTD抗白血病机制。方法采用悬浮培养对数生长期细胞,研究Jurkat细胞株在NTCD作用下的体外机制:实验分成两组:(1)对照组,培养液中不添加任何药物;(2)NCTD组,配制20 mg/L浓度的NTCD作用于Jurkat细胞系。两组细胞共同培养72 h。运用Real-time PCR技术分别对NTCD作用前后的Jurkat细胞株中的p53及survivin mRNA表达进行定量检测。应用t检验进行统计分析。结果 NCTD组p53 mRNA表达较对照组细胞株上升3%,相反survivin mRNA较对照组下降20%,应用统计学方法每两组进行比较均有统计学意义(P<0.01)。结论去甲斑蝥素上调抑癌基因p53 mRNA,下调癌基因survivin mRNA途径可能是NCTD抗白血病的主要机制之一。Objective To investigate the mechanism of norcantharidin (NCTD) against the acute lymphocytic leukemia. Methods The conventional suspention cell culture method was used for cultivating the Jurkat cell lines in vitro. The cells were categorized into two groups based on treatment. (1)The control group was treated with medium only. (2)The NCTD group was treated with NTCD (20 mg/L). Both cells were cultivated for 72 h. The mRNA gene expression of survivn and p53 were compared between groups by RT-PCR. The student t-test was used for statistical analysis. Results The gene expression of p53 was significantly increased by 3% compared with control group (P 〈0. 01 ). However, the survivin gene showed a 20% reduction (P 〈 0. 01 ). Conclusion NTCD might play a critical role in regulating the gene expression of survivin and p53, which contributes to the resistant to acute lymphocytic leukemia.
