摘  要:目的建立酵母多糖(Zym)致全身炎症反应综合征(SIRS)大鼠模型,观察大鼠的肠屏障功能变化并探讨SIRS大鼠肠屏障功能障碍的机制及治疗。方法腹腔注射Zym-石蜡混悬液制备大鼠SIRS模型,分为Zym750、500、250、125mg/kg模型组,观察各组大鼠体温、呼吸、心率、血白细胞计数,造模24h后按照SIRS大鼠模型诊断标准评价模型是否成功,同时设立空白对照组。检测造模后血浆D-乳酸、二胺氧化酶(DAO)、24h尿甘露醇/乳果糖(L/M)评估大鼠肠屏障功能,检测血肿瘤坏死因子-α(TNF—α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、内毒素(ET)水平了解炎症情况。对各组大鼠末端回肠行H—E染色以分析肠绒毛损伤情况。结果Zym500mg/kg组、Zym250mg/kg组大鼠血浆D-乳酸、DAO、24h尿液L/M与空白对照组比较差异有统计学意义(P〈0.05或P〈0.01);Zym500mg/kg组与Zym250mg/kg组比较差异有统计学意义(P〈0.05)。Zym500mg/kg组、Zym250mg/kg组大鼠血液TNF—α、IL-1β、IL-6、IL-10、ET与空白对照组比较差异有统计学意义(P〈0.05或P〈0.01);Zym500mg/kg组与Zym250mg/kg比较差异有统计学意义(P〈0.05或P〈0.01)。Zym500mg/kg组、Zym250mg/kg组大鼠与空白对照组比较肠黏膜绒毛变短、隐窝深度变浅,差异有统计学意义(P〈0.05);Zym500mg/kg组与Zym250mg/kg组比较差异有统计学意义(P〈0.05或P〈0.01)。结论酵母多糖致SIRS大鼠模型肠屏障功能发生改变。Objective To establish a model of systemic inflammatory response syndrome (SIRS) induced by zymosan (Zym) in rats, observe the changes of intestinal barrier function of rats, and explore the mechanism of SIRS in rats and treatment strategies. Methods The rat model of SIRS was established by intraperitoneal injection of a suspension of Zym and paraffin, with Zym concentrations of 750 mg/kg, 500 mg/kg, 250 mg/kg and 125 mg/kg. Meanwhile, the body temperature, respiratory rate, heart rate, and blood leukocyte count of the rats were recorded. Then, 24 h after modeling, the resultant model was assessed according to the standards for diagnosis of organ dysfunction syndrome in modeled rats with SIRS. Meanwhile, a blank control group was set up. The levels of plasma D - lactic acid, diamine oxidase ( DAO), 24 h urine lactulose/mannitol ratio (L/M) were measured after modeling to evaluate the intestinal barrier functions of the rats. Furthermore, the amounts of blood tumor necrosis factor α( TNF - α), interleukin - 1β ( IL - 1β), interleukin-6 (IL-6), interleukin- 10 (IL- 10), and endotoxin (ET) were determined to evaluate the inflammatory conditions of the rats. The terminal ileum of each rat was adopted to analyze intestinal villus injury through H - E staining. Results Compared with the blank control group, the Zym 500 mg/kg group and 250 mg/kg group showed remarkably different levels of plasma D - lactic acid, DAO, 24 h L/M (P 〈 0.05 or P 〈 0.01 ). Compared with the Zym 250 mg/kg group, the Zym 500 mg/kg group also presented significant difference ( P 〈 0.05). Furthermore, the Zym 500 mg/kg group and 250 mg/kg group showed remarkably different quantities of blood TNF - α, IL - 1β, IL - 6, IL - 10, and ET, compared with the blank control group (P 〈 0.05 or P 〈 0.01 ).
