摘 要:目的探讨甲基苄基亚硝胺(MBNA)致食管癌前病变时大鼠血清氧化还原水平的变化及鸦胆子油的影响。方法Wistar大鼠按3.5mg/kg皮下注射MBNA溶液,每周2次,维生素C组自诱变首日起皮下注射,每周5次,连续10周,治疗4周后处死所有大鼠,检测总SOD活性、总谷胱甘肽水平和总抗氧化能力并观察食管黏膜变化。结果各组瘤体数较模型组有所减少(P<0.01),除维生素C组外,各组瘤体体积均无差异(P>0.05);模型组较正常组总SOD活性,总谷胱甘肽水平和总抗氧化能力有所升高(P<0.05);鸦胆子油组较模型组降低(P<0.01)。结论经鸦胆子油治疗后,总SOD活性,总谷胱甘肽水平和总抗氧化能力降低,增强肿瘤组织的氧化损伤可能是鸦胆子抗肿瘤的机制之一。Objective To investigate the changes of serum redox level in precancerous esophageal lesions in Wistar rats following treatment with methyl benzyl nitrosamines (MBNA) and Java brucea fruit oil. Methods Wistar rats with precancerous esophageal legions were injected with MBNA subcutaneously (3.5 mg/kg, twice a week) for 10 weeks, while control rats were injected with vitamin C (5 times a week) for 10 weeks. Rats were treated with Java brucea fruit oil for4 weeks, prior to sacrifice in week 14. Subsequently, we determined the levels of glutathione, antioxidant capacity, SOD activity and pathological changes in esophageal mucosa for both treatment and control groups. Results Rats treated with Java brucea fruit oil had a significantly decreased number of tumor lesions compared with the control group (P 〈 0.01 ). There were no differences in tumor number between rats treated with MBNA and vitamin C (P 〉 0.05 ). There was an obvious increase in the level of glutathione, total antioxidant capacity and total SOD activity in the rats treated with MBNA compared to the control group ( P 〈 0. 05 ), which was significantly decreased fol- lowing Java brucea fruit oil treatment (P 〈 0.01 ). Conclusion Java brucea fruit oil treatment can effectively reduce the levels of glutathione, antioxidant capacity and SOD activity, and is therefore a potential antitumor drug which acts via inducing oxidative damage in tumor tissues.
