摘  要:目的:探讨细胞增生与凋亡蛋白在结膜松弛症手术切除组织中的表达情况及其所起的作用。方法收集结膜松弛症30例(30只眼)手术切除标本及正常对照组结膜组织15例(15只眼)。采用免疫荧光的方法比较结膜松弛症组及正常组结膜中Bcl-2相关X蛋白基因( Bax)、半胱氨酸天冬氨酸蛋白酶-3(caspase-3)、B细胞淋巴瘤/白血病-2基因(Bcl-2)以及增生细胞核抗原(PCNA)的表达情况。结果结膜松弛症结膜组织中Bcl-2阳性表达面积为(741.30±63.24)μm2,正常结膜中为(728.70±71.66)μm2;结膜松弛症结膜组织中Bax阳性表达面积为(859.20±81.16)μm2,正常结膜中为(722.60±70.51)μm2;结膜松弛症结膜组织中PCNA阳性表达面积为(457.60±94.64)μm2,正常结膜中为(411.20±81.25)μm2;结膜松弛症结膜组织中 caspase-3阳性表达面积为(881.70±41.38)μm2,正常结膜中为(698.90±56.66)μm2,Bax及caspase-3在结膜松弛症结膜组织中表达的荧光强度明显强于正常结膜组织(P〈0.05),而两组间Bcl-2及PCNA表达的荧光强度无明显差异(P〉0.05)。结论结膜松弛症结膜组织中凋亡蛋白的表达增强提示细胞凋亡可能是结膜松弛症发生过程中的重要环节。Objective To investigate the expression and function of cell proliferation and apoptosis protein in conjunctival tissue excised from conjunctivochalasis ( CCh ) . Methods Thirty speciments of conjunctival tissue excised from CCh and 15 normal conjunctival tissues were collected. The expression of Bax 、caspase-3、Bcl-2 and PCNA in each group was detected by immunofluorescence. Results The positive cells area of Bcl-2 in conjunctival tissue of CCh was (741. 30 ± 63. 24) μm2, and that was (728. 70 ± 71. 66) μm2 in normal tissue. The positive cells area of Bax in conjunctival tissue of CCh was (859. 20 ± 81. 16) μm2, and that was (722. 60 ± 70. 51) μm2 in normal tissue. The positive cells area of PCNA in conjunctival tissue of CCh was (457. 60 ± 94. 64) μm2, and that was (411. 20 ± 81. 25) μm2 in normal tissue. The positive cells area of caspase-3 in conjunctival tissue of CCh was (881. 70 ± 41. 38) μm2, and that was (698. 90 ± 56. 66) μm2 in normal tissue. The average fluorescent intensities of Bax and caspase-3 in the conjunctival tissue of CCh were significantly higher than those in normal tissue (P 〈0. 05). No statistically sighficant difference was found in the average fluorescent intensity of Bcl-2 and PCNA between CCh group and normal group (P〉0. 05). Conclusion The enhancement of apoptosis protein expression in 〈br〉 conjunctival tissue of CCh suggests that apoptosis may be an important part in the process of the occurrence of conjunctivochalasis.
