摘 要:目的探讨Wnt信号通路中关键蛋白Wnt3a和TCF4在卵巢癌中的表达水平以及临床意义。方法 ELISA法检测10例卵巢癌患者手术前后和10例健康对照者血浆Wnt3a浓度,Western blot、qRT-PCR法检测卵巢癌患者与卵巢良性病变者的卵巢组织Wnt3a、TCF4水平。采用Western blot法检测卵巢癌细胞株SKOV3与HO8910中TCF4的表达,采用dn TCF4质粒转染技术获得TCF4表达下降的SKOV3/dn TCF4细胞,使用细胞划痕实验比较卵巢癌细胞转移能力。结果卵巢癌患者血浆Wnt3a水平高于健康对照者(P<0.05),且术后血浆Wnt3a水平低于术前(P<0.05);卵巢癌组织中Wnt3a、TCF4水平高于卵巢良性病变者(P<0.05);SKOV3的TCF4表达量高于HO8910且转移潜能也明显高于后者(P<0.05);将SKOV3的TCF4敲低,其转移能力明显减弱(P<0.05)。结论 Wnt信号通路参与卵巢癌的发生发展,Wnt3a和TCF4有可能成为监测卵巢癌进展的临床指标之一。Objective To investigate the expression levels and clinical significance of Wnt3 a and TCF4 belonging to Wnt signaling pathway in ovarian cancer. Methods ELISA was used to detect plasma Wnt3 a concentration in10 ovarian cancer patients before and after surgery and 10 healthy controls. Western blot and qRT- PCR were used to detect Wnt3 a and TCF4 levels in ovarian cancers and benign ovarian lesions. The expression of TCF4 in ovarian cancer cell lines SKOV3 and HO8910 was detected by Western blot method. Dominant negative TCF4( dn TCF4)plasmid was stably transfected to obtain TCF4 knockdown cell line SKOV3/dn TCF4. Wound healing assay method was used to compare the metastasis of ovarian cancer cell lines. Results Plasma Wnt3 a levels in ovarian cancer patients were averagely higher than those in healthy controls( P〈0. 05). The Wnt3 a levels after surgery were lower than those before surgery for the ovarian cancer patients( P〈0. 05). The Wnt3 a and TCF4 expressions in ovarian cancer tissues were higher than those in benign ovarian lesions( P〈0. 05). TCF4 expression in SKOV3 with high metastatic potential was higher than that in HO8910 with low metastatic potential( P〈0. 05). TCF4 knockdown in SKOV3/dn TCF4 resulted in a significantly reduced metastasis( P〈0. 05). Conclusion Wnt signaling pathway is involved in the development of ovarian cancer,and Wnt3 a and TCF4 may become useful clinical indicators for monitoring the progress of ovarian cancer.
