摘 要:扩展青霉(Pennicilum expansum)是导致苹果采后青霉病的主要致病菌,为探讨Bacillus amyloliquefaciens BA-16-8所产生的脂肽类抗生素丰原素(Fengycin)对Penicillum expansum呼吸代谢、线粒体复合酶活性和营养物质利用的影响。通过溶氧量测定实验检测Fengycin处理对P.expansum的呼吸抑制率,通过线粒体酶活力测定实验检测Fengycin处理对P.expansum线粒体复合酶活力的影响,通过DNS比色法和双缩脲法测定Fengycin处理对P.expansum利用总糖和总蛋白的能力。结果表明,Fengycin主要抑制病原菌呼吸代谢的TCA途径。Fengycin是通过结合P.expansum线粒体复合酶II和III的相关基因,从而使该基因的表达受阻,进而影响线粒体酶的活性。随着Fengycin处理浓度的增加,P.expansum对总糖和总蛋白质的吸收及利用能力显著下降。研究结果表明,Fengycin的作用可影响P.expansum的呼吸,从而降低其代谢水平,最终抑制病原菌的生长。Penicillum expansum is considered as the main pathogen that causes blue mold in postharvest apples. The effect of fengycin produced by Bacillus amyloliquefaciens BA-16-8 on the respairation, mitochondria complexes activity and nutrients utilization of P. expansum was discussed by soluble oxygen experiment, DNS and biuret method. As was shown in the results, fengycin mainly restrainedTCA pathway. Fengycin inhibit the mitochondrial complexes II and III enzymatic activity by combining with mitochondrial enzyme genes. Moreover, fengycin conld inhibit the growth of mycelium by reducing the utilization of carbohydrates and protein in P. expansum. To sum up, fengycin could inhibit the respiratory metabolism and reduce the biochemical metabolism level in P. expansum and finally caused the growth inhibition.
