miR-34a在牙龈鳞状细胞癌组织中的表达及其临床意义

摘　　要：目的:检测牙龈鳞状细胞癌(GSCC)及癌旁组织中小分子RNA 34a(miR-34a)的表达水平,探讨miR-34a表达与GSCC患者肿瘤大小、恶性程度以及患者年龄、性别、肿瘤的部位及有无淋巴结转移等临床病理特征之间的关系。方法:收集30例确诊为GSCC患者手术切除的GSCC组织及相应癌旁组织。采用荧光实时定量PCR(qRT-PCR)法检测GSCC和癌旁组织中miR-34a的表达水平,比较不同临床病理特征GSCC患者GSCC组织中miR-34a的表达水平。结果:与癌旁组织比较,GSCC组织中miR-34a的表达水平明显降低(t=3.03,P=0.004);GSCC组织中miR-34a表达水平与GSCC患者肿瘤大小及恶性程度有关联(P<0.05),但miR-34a表达水平与患者的年龄、性别、肿瘤所在部位及有无淋巴结转移等无关联(P>0.05)。结论:GSCC患者肿瘤组织中miR-34a表达水平明显降低,检测miR-34a表达水平有助于判断GSCC患者的病理分型。Objective：To detect the expression levels of microRNA-34a（miR-34a）in gingival squamous cell carcinoma（GSCC）tissue and adjacent tissue,and to clarify its correlation with the pathological characteristics of GSCC patients,including tumor size,malignancy degree,age,gender,tumor site and lymph node metastasis.Methods：The tissue samples including 30 GSCC tissues and matched carcinoma adjacent tissues were selected.The expression levels of miR-34 ain GSCC tissue and carcinoma adjacent tissue were detected by quantitative reverse transcription-PCR（qRT-PCR）method.The relationship between its expresion level and the clinicopathological characteristics of GSCC patients were analyzed.Results：Compared with carcinoma adjacent tissue,the expression level of miR-34 ain GSCC tissue was significantly decreased（t=3.03,P =0.004）.The expression level of miR-34 ain GSCC tissue had no relationship with the age,gender,and lymph node metastasis of the patients（P〉0.05）,but had relationship with the tumor size and tumor malignancy degree（P 0.05）.Conclusion：The expression level of miR-34 ais down-regulated significantly in GSCC tissue.The detection of the expression level of miR-34 acontributes to judge the histological subtype.
