摘 要:目的了解不同剂量率X线照射对非小细胞肺癌细胞周期的影响,以期为临床制定放疗计划提供一定的实验依据。方法选取A549肺癌细胞进行培养48 h,采用直线加速器行X线照射,照射剂量为6 Gy,照射剂量率分别为1、2、4、6 Gy/min,照射后24 h收集细胞,用70%乙醇固定,RNA酶处理,碘化丙啶染色,采用流式细胞学的方法检测细胞周期。结果 X线照射后细胞周期分布发生明显的改变,细胞在G2/M期发生明显阻滞,S期的细胞明显减少,其中以2 Gy/min照射的细胞G2/M期的阻滞最为明显。结论 X线照射影响A549细胞周期的进程,相同的剂量,不同的剂量率对细胞周期的影响存在明显差异。选取最佳剂量率进行治疗,可能有助于提高肿瘤放射治疗效果。Objective To investigate the effects of dose rate of X-ray on cell cycle of non-small-cell lung cancer (NSCLC) cells. Method A549 cells of a NSCLC cell line were cultured for 48 hours and then irradiated with X-rays at dose of 6 Gy from a linear accelerator. The dose rates were 1, 2, 4 and 6 Gy/min, respectively. Cells were collected 24 hours after irradiation, and then were fixed with 70%ethanol, treated with RNase and stained with propidium iodide. Cell cycle distribution was analyzed using BD FACSCalibur. Result Cell cycle distribution in irradiated cells was altered sig-nificantly. Radiation induced cell cycle arrest in G2/M phase and cells decreased in S phase. The G2/M phase arrest was most significant in cells irradiated at dose rate of 2 Gy/min. Conclusion X-ray irradiation influences the cell cycle distri-bution of A549 cells. Though at same radiation dose, different dose rates impose varied effects on cell cycles. Thus, treat-ment with optimal dose rate would improve the efficacy of radiotherapy.
