摘　　要：目的:紫芝是多孔菌科灵芝属的重要传统中药。细胞色素P450是一类含血红素的单加氧酶。本研究的目的是对紫芝基因组编码的CYP450序列进行比对及系统进化分析。方法:发掘紫芝基因组编码的所有P450基因,构建紫芝CYP450系统进化树,比较灵芝属3个已测序物种中P450基因家族的差异,并对紫芝中可能参与三萜生物合成的P450基因家族进行染色体定位和内含子相位分析。结果:紫芝基因组共编码228个CYP450基因,其中包括9个假基因,在进化上分为3个分支,分布在41个家族中,其中CYP5359家族最大,包含有53条序列。紫芝所有的P450基因家族在其他两个灵芝属真菌中都存在。CYP512和CYP5144家族可能参与灵芝酸生物合成,其中一些家族成员可能是通过基因复制进化而来的。结论:紫芝P450基因家族的系统进化分析与进化机制研究为进一步揭示紫芝P450基因的功能与生理生化作用奠定了基础。Objective： Ganoderma sinense,belonging to the Polyporales,Ganodermae,is one of the most wellknown medicinal mushrooms in the world. Cytochrome P450 s are monooxygenases containing heme. Our study aims to understand the evolution of P450 families in G. sinense. Methods： We found all P450 genes from the G. sinense genome and constructed their phylogenetic tree. The P450 s from G. sinense were compared with those from the other two sequenced fungi from Ganoderma genus. We also analyzed the chromosome location and intron phases of the P450 genes possibly involved in the biosynthesis of triterpenoids. Results： The G. sinense genome encoded a total of 228 P450 genes,including 9 pseudogenes. All of them were divided into three clans and 41 families, among which, CYP5359 is the biggest family containing 53 members. All the P450 families in G. sinense also exist in the other two sequenced Ganoderma species. CYP512 and CYP5144 families were supposed to be involved in the biosynthesis of triterpenoids and some of their members were generated by gene duplications. Conclusion： We analyzed the phylogenetics of P450 s in G. sinense and explored their possible evolutional mechanism,which will lay the foundation for further study on the function and physiological activities of these P450 s.
