摘  要:目的探讨Toll样受体4(TLR4)、髓样分化因子88(MyD88)在高脂高糖饮食诱导的非酒精性脂肪性肝病(NAFLD)大鼠肝组织中的表达及其意义。方法选取SD大鼠20只,随机分为两组,模型组SD大鼠给予高脂高糖饲料喂养,正常组给予普通饲料喂养,10周末分别进行如下检测:1计算肝指数;2HE染色,光镜下观察各组大鼠肝组织的病理改变;3应用RT-PCR法检测大鼠肝组织TLR4、MyD88 mRNA的表达。结果模型组的肝指数较正常组明显增加(P<0.01);模型组的NAFLD活动度计分与正常组比较差异有统计学意义(P<0.01);模型组大鼠肝组织TLR4和MyD88的mRNA表达水平与正常组比较差异有统计学意义(P<0.01)。结论持续10周的高脂高糖饮食喂养可以诱导NAFLD大鼠模型,模型组大鼠肝组织TLR4 mRNA和MyD88 mRNA表达升高,在肝细胞炎症改变中起重要作用。Objective To investigate the expression and significance of TLR4 and MyD88 in liver tissues of rats with nonalcoholic fatty liver disease(NAFLD) induced by high-fat and high-sugar diet. Methods Twenty SD rats were selected and randomly assigned into two groups. The SD rats in the model group were fed with high-fat and high-sugar feed, and the normal group was fed with normal feed. The following tests were carried out at the end of 10 weeks: ①liver index was calculated; ②HE staining was performed. The pathological changes of liver tissues were observed under light microscope; ③The expression of TLR4 and MyD88 mRNA in liver tissues were detected by RT-PCR. Results The liver index in the model group was significantly higher than that in the normal group(P〈0.01); the NAFLD activity score in the model group was statistically significantly different from that in the normal group(P〈0.01); the mRNA expression levels of TLR4 and MyD88 in rats" liver tissue in the model group were also statistically significantly different from those in the normal group(P〈0.01). Conclusion High-fat and high-sugar diet fed for consecutive 10 weeks can induce increased expression of TLR4 mRNA and MyD88 mRNA in the liver tissue of NAFLD rats in the model group, which plays an important role in the inflammatory changes of liver cells.
