摘 要:目的应用核磁共振(NMR)波谱学的代谢组学方法,检测中医体质分类中阳虚体质与平和体质的血浆代谢物,研究阳虚体质中特征代谢物的特点。方法收集阳虚体质与平和体质各8例的血液样品,采用横向豫驰时间的分布曲线磁共振脉冲序列(CPMG)采集血液样品数据,观测血浆中的小分子代谢物。用多元统计分析方法比较阳虚体质组与平和体质组血浆内源性差异代谢产物。结果血浆中的代谢产物,与平和体质组相比,在阳虚体质组中乳酸(shift 1.33;t=-3.018)、低密度胆固醇脂蛋白(shift 1.26;t=-2.849)、脂肪酸(shift 2.25;t=-3.325)、氧化三甲胺(shift 3.26;t=-5.217)、N-乙酰糖蛋白(shift 2.03;t=-4.684)水平都有不同程度下降;而磷脂酰胆碱(shift 3.22;t=3.901)、葡萄糖(shift 3.42;t=3.390)与谷氨酰胺(shift 2.12;t=5.177)水平都有不同程度升高。结论阳虚、平和体质不仅存在体内生物合成与分解中能量代谢、糖代谢、脂代谢、氨基酸代谢等的差异,也存在神经递质、脏腑功能的改变。Objective Plasma etabonomics was used to study yang-deficiency constitution, for analyzing the characteristic metabolites. Methods Plasma samples from 8 participants with yang-deficiency constitution and 8 participants with balanced constitution were collected. Carr-Purcell-Meiboom-Gill(CPMG) pulse sequence was used to collect plasma data,and to observe small molecule metabolites in plasma. Differences in endogenous metabolites were detected by multivariate statistical analysis. Results Compared with balanced constitution,lactic acid(t=3. 018), low density lipoprotein cholesterol(t=-2. 849),fatty acids (t=- 3. 325),trimethylamine oxide(t=-5. 217), N - acetyl glycoproteinare(t=- 4. 684) lower in plasma of yang-deficiency con- stitution,while glucose(t=3. 390) ,phosphatidyl choline(t=3. 901) and glutamine(t=5. 177) are higher. Conclusion The biomarkers supply new ways for the individual differences from the point of view of metabonomics. Yang-deficiency constitution has some metabolism disorder of energy, sugar, lipid, and amino acid metabolism, the neurotransmitter, and viscera function.
