摘  要:本试验旨在探讨β-胡萝卜素对小鼠肠黏膜上皮紧密连接蛋白的保护作用。将40只体重在22~25 g的雄性健康昆明小鼠,随机分为4组,分别为对照组、β-胡萝卜素处理组、脂多糖(LPS)组、保护组,每组10只,对照组饲喂正常鼠粮,β-胡萝卜素组饲喂含有50 mg/kgβ-胡萝卜素的鼠粮,LPS组饲喂正常鼠粮并每天注射10 mg/kg LPS,保护组饲喂含有50 mg/kgβ-胡萝卜素的鼠粮并每天注射10 mg/kg LPS。结果表明:与对照组相比,LPS的注射能显著降低小鼠血液中IL-2、IL-4含量(P〈0.05),显著提高小鼠血液中NO含量(P〈0.05),影响小鼠结肠结构并显著降低结肠紧密连接蛋白ZO-1的蛋白表达(P〈0.05);而与LPS组相比,保护组的小鼠血液中IL-2、IL-4含量以及结肠紧密连接蛋白ZO-1的蛋白表达显著升高(P〈0.05),NO含量显著降低(P〈0.05)。结果显示,β-胡萝卜素可对由LPS应激造成的小鼠肠黏膜损伤具有明显的保护作用,该结果对保护肠屏障功能具有重要意义。The present study was conducted to investigate the protective effects of B-carotene on tight junction proteins of mouse intestinal epithelium. Fourty healthy male mice weighing 22-25 g were randomly divided into four groups (n=10), namely the control group, β-carotene treatment group, LPS group, protecting groups. Control group was fed with normal diet, β-carotene group was fed with normal diet containing 50 mg/kg β-carotene, LPS group was fed with normal diet and daily injections of 10 mg/kg LPS, the protection group was fed with diet containing 50 mg/kg β- carotene and injecting 10 mg/kg LPS every day. The results showed that compared with control group, LPS injection could significantly reduce IL-2, IL-4 content in the blood of mice (P〈0.05), significantly increase NO levels in the blood of mice (P〈0.05), structural damage of colon of mice and significantly reduced colon expression of tight junction protein ZO-1 (P〈0.05). junction protein ZO-1 in decreased(P〈0.05). These Compared with LPS group, IL-2, IL-4 content in blood of and expression of colon tight protection group were significantly increased, and NO content in blood was significantly results indicated that B-carotene could protect intestinal mucosa under LPS stress and these results are important for the protection of intestinal muncosa function.
