摘  要:探究蓝藻PCC7002是否可以对黄芩苷进行生物转化。通过吸光度法检测蓝藻生长能力,利用薄层色谱法检测转化产物,利用高效液相色谱法进一步确认。确定蓝藻生物转化方法为:先将蓝藻培养到浓度达到A700 nm值0.8,再将培养液p H调为5.5,加入0.1 mg/m L的底物,在35℃摇床培养3 d后,取上清进行薄层色谱检测,结果显示薄层板上有新的点出现,经高效液相验证为新的转化产物,极性略大于黄芩苷。蓝藻PCC7002能对黄芩苷进行转化,转化效率初步推断为37.5%。To explore whether Cyanobacteria PCC7002 has the ability to biotransform Baicalin to other compounds.The conditions for the biological transformation system were optimized.The growth of the Cyanobacteria was measured by absorptiometry at 700 nm.Baicalin was stable when the MA culture medium p H4.5 or 5.5,but not stable at p H 6.8 which is the most suitable for the cyanobacteria cells growth.The temperature had no significant effect on the stability of baicalin both at 30 °C and 35 °C.The latter is much suitable for the cells growth.Therefore,the cyanobacteria cells were firstly cultured in MA medium(p H 6.8) at 35℃ until the cells concentration reached to A700nm= 0.8.Then the p H of the medium was adjusted to 5.5 and the baicalin was added into the medium.The temperature was kept at 35 ° C.The concentration of substrate was 0.1 mg / m L.The Cyanobacteria was cultured for3 days in incubator shaker with 120 r / min after the substrate added.Then the supernatants were collected after 3 days biotransformation.The supernatants were analyzed by the thin layer chromatography(TLC).The developing agent consisted of ethyl acetatebutanone-formic acid-methanol(10∶ 7∶ 0.5∶ 1) and colored under the UV.There was a new point on the thin plate with a smaller migration rate.The supernatants were also tested by the high performance liquid chromatography(HPLC).The condition of HPLC were as follows: the mobile phases consisted of methanol-water-phosphonic acid(47∶ 53 ∶ 0.2); the UV detector at 280 nm; Speed,1 m L / min; Column temperature,40 °C.A new peak was displayed on the chromatography which appeared earlier than the substrate.In conclusion,baicalin could be bio conversed to other components by cyanbacteria PCC7002.The conversion efficiency was 37.5%.What can be preliminarily determined is that the transformation products had higher polarity than baicalin.
