摘 要:目的研究胃癌和癌旁组织中Aurka mRNA及其蛋白的表达,并分析胃癌组织中Aurka mRNA表达水平与患者临床病理学特征的关系。方法回顾性收集2011年4月至2013年9月期间于川北医学院附属医院胃肠外科接受手术的198例胃癌患者的手术标本,包括癌组织和癌旁组织。采用实时荧光定量逆转录聚合酶链反应(RT-PCR)和免疫组织化学染色法分别检测胃癌和癌旁组织中Aurka mRNA及其蛋白的表达,并进行比较;同时探讨胃癌组织中Aurka mRNA表达水平与患者临床病理学特征的关系。结果 RT-PCR结果显示,胃癌组织与癌旁组织中Aurka mRNA的表达水平分别为16.62±1.85和7.10±1.59,癌组织较高,差异有统计学意义(t=-38.58,P<0.05);免疫组织化学染色结果显示,胃癌组织中Aurka蛋白的表达阳性率高于癌旁组织〔93.9%(186/198)比16.2%(32/198),P<0.01〕。胃癌组织中Aurka mRNA的表达水平与性别、年龄、肿瘤位置、术前癌胚抗原(CEA)和CA19-9表达均无关(P>0.05),而与肿瘤TNM分期、T分期、N分期及分化程度均有关(P<0.05)。幽门螺杆菌(HP)阳性患者胃癌组织中Aurka mRNA的表达水平为15.38±1.73,高于HP阴性患者(7.20±1.86),差异有统计学意义(t=-3.74,P<0.01)。结论 Aurka基因及其蛋白在胃癌组织中的表达水平均高于癌旁组织,提示其作为一种致癌基因,在胃癌的发生和发展过程中扮演着重要角色,可作为胃癌患者预后及复发的预测指标。Objective To investigate the expressions of Aurka mRNA and its protein in gastric cancer and adjacent tissues, and to analyzed the relationship between the expression level ofAurka mRAN and clinicopathological characteristics of gastric cancer patients. Methods Retrospectively collected the gastric cancer and adjacent tissues of 198 gastric cancer patients who treated in the Affiliated Hospital of North Sichuan Medical College between April 2011 and September 2013. Using real time quantitative reverse transcription polymerase chain reaction (RT-PCR) method and immunohistochemical staining to detected expressions of Aurka mRNA and its protein of gastric cancer and adjacent tissues respectively. At the same time explored the relationship between expression level of Aurka mRNA in gastric cancer tissues and the clinicopathological features of gastric cancer patients. Results RT-PCR results showed that, the expression level of Aurka mRNA in gastric cancer tissues was 16.62±1.85, which was significantly higher than that of adjacent tissues (7.10±1.59), P〈0.05. Immunohistochemistry results showed that, the positive rate of Aurka protein in gastric cancer tissues was obviously higher than of adjacent tissues [93.9% (186/198) vs. 16.2% (32/198), P〈0.01]. The expression level of Aurka mRNA in gastric cancer tissues was significantly related with a part of the clinicopathological parameters, including TNM staging, T staging, N staging, and differentiation (P〈0.05), but there was no relationship with gender, age, location, and carcinoembryonic antigen (CEA)/CA19-9 level in peripheral blood (P〉0.05). The expression level ofAurka mRNA in Helicobacter pylori (HP)-positive gastric cancer patients was higher than that of HP-negative gastric cancer patients (15.38±1.73 vs. 7.20±1.86, t=-3.74, P〈0.01).
