摘  要:为了检测吞噬和运动蛋白家族中的ELMO3在结肠癌组织中的表达情况,并探讨其与结肠癌临床病理参数间的相关性及意义,本研究应用免疫组化法检测了组织芯片中75例结肠癌及相匹配的癌旁正常结肠组织中ELMO3蛋白的表达水平。结果显示,ELMO3在结肠癌组织中的阳性表达率显著高于对应的癌旁组织(P<0.001);不同性别、年龄和肿瘤部位的ELMO3表达水平均无显著性差异(P>0.05);不同肿瘤大小、结肠癌分化程度、浸润深度和远处转移的结肠癌组织中的ELMO3表达水平存在显著性差异(P<0.05)。且ELMO3的表达与肿瘤分化程度、淋巴结转移和临床TNM分期均呈正相关性(R=0.386、R=0.608和R=0.760,P<0.05),即随着肿瘤分化程度、淋巴结转移和临床分期的增高,ELMO3的表达水平也增高。尤其是ELMO3在发生淋巴结转移的结肠癌组织中细胞染色强度显著高于未发生淋巴结转移的结肠癌组织中细胞染色强度,其差异具有统计学意义(P<0.05)。这些结果表明,ELMO3在结肠癌组织中高表达,其表达与肿瘤分化程度、淋巴结转移和临床TNM分期相关,提示ELMO3表达水平与结肠癌生物学行为密切相关,有望成为结肠癌侵袭转移治疗的有效靶点。To detect the expression of ELMO3 in colon cancer and to explore its association with the clinicopathological characteristics,immunohistochemistry for ELMO3 was conducted on tissue microarray containing 75 cores of colon cancer and 75 cores of normal colon tissues matched to tumor sections.The expression level of ELMO3 was significantly higher in colon cancer tissues than that in normal adj acent colon tissues (P〈0.001).ELMO3 expression was not related to any specific patient characteristics,such as age,gender and tumour location (P〉0.05 ),whereas it was correlated with the grade of colon cancer differentiation,the depth of invasion,lymph node metastasis,distant metastasis and TNM classification.In addition,there was positive correlation between ELMO3 expression level with the grade of colon cancer differentiation,lymph node metastasis and clinical TNM stages (R=0.386,R=0.608 and R=0.760,P〈0. 05).As the degree of tumor differentiation,lymph node metastasis and clinical TNM staging increased,the expression level of ELMO3 was higher.Stronger staining of ELMO3 was detected in cancer cells with lymph node metastasis group than cancer cells without lymph node metastasis (P〈0.05).In conclusion, ELMO3 is up-regulated in colon cancer.Specifically,it is of clinical relevance to differentiation,lymph node metastasis and clinical TNM staging of colon cancer,and its expression might be closely related to clinical biological behaviors.ELMO3 will be expected to become an effective target for biotherapy for metastatic colon cancer.
