宫颈癌组织中胰岛素样生长因子的表达及意义

摘　　要：目的探讨胰岛素样生长因子IGF-2在不同期别宫颈癌中的表达及其意义。方法收集诸城市妇幼保健院和青岛大学附属医院在2013年1月-2014年2月间收治的不同期别宫颈癌石蜡标本70例,宫颈上皮内瘤变蜡块组织28例,同期因良性疾病行子宫切除术的宫颈蜡块标本20例作为对照。全部病例经病理诊断证实。采用免疫组织化学SP法检测IGF-2在不同组织中的表达。使用SPSS13.0统计软件包对数据进行处理,所有计数资料使用卡方检验。统计学意义的差异判断标准为P<0.05。结果 IGF-2蛋白主要位于胞质表达。IGF-2蛋白在宫颈癌组织、宫颈上皮内瘤变组织、正常宫颈组织的表达百分率依次递减,分别为68.57%(48/70)、28.57%(8/28)、10.00%(2/10),正常宫颈组织与宫颈癌组织、宫颈上皮内瘤变组织比较,差别有统计学意义(P<0.05),正常宫颈组织与宫颈上皮内瘤变组织比较,差异有统计学意义(P<0.05)。结论宫颈正常组织中IGF-2低表达或者不表达,宫颈上皮内瘤变不同期别组织中呈现中等量表达,在不同分期的宫颈癌组织中表达量较高。Objective To explore the expression and significance of insulin- like growth factor- 2（ IGF- 2） in cervical cancer of different stages. Methods Seventy paraffin- embedded cervical cancer specimens of different stages were collected from Zhucheng Maternal and Child Health Care Hospital and Affiliated Hospital of Qingdao University from January 2013 to February 2014,including 28 specimens of cervical intraepithelial neoplasia（ CIN）; 20 paraffin- embedded cervical specimens after hysterectomy because of cervical benign diseases were collected as control group; all the specimens were confirmed by pathological examination. Immunohistochemical SP method was used to detect the expressions of IGF- 2 in different groups. SPSS 13. 0 software was used for statistical analysis,all the enumeration data was analyzed by chi- square test,the significant level was 0. 05. Results IGF- 2 protein mainly expressed in the cytoplasm. The expression percentages of IGF- 2 in cervical cancer,CIN,and normal cervical tissue decreased gradually,which were 68. 57%（48 /70）,28. 57%（8 /28）,and 10. 00%（2 /10）,respectively. There was statistically significant difference in IGF- 2 protein expression beween normal cervical tissue and cervical cancer,CIN（ P〈 0. 05）; there was statistically significant difference beween normal cervical tissue and CIN（ P〈 0. 05）. Conclusion IGF- 2 rarely expresses in normal cervical tissue,moderately expresses in CIN,and highly expresses in cervical cancer of different stages. IGF- 2 can promote the development of cervical cancer.
