摘 要:目的 运用尖端放电喷雾电离质谱技术鉴定肺癌差异性磷脂。方法 在不对肺组织进行任何预处理的前提下,采用自制的甲醇萃取辅助的尖端放电喷雾离子源对14例肺癌患者的肺癌组织和癌旁组织的差异性磷脂[质荷比(m/z)756.7]及标准品[二棕榈酰磷脂酰胆碱(DPPC)+Na+]进行多级质谱检测,萃取剂甲醇的流速为4μL/min,设置离子源为正离子模式,一级质谱扫描范围为m/z 750~850,多级谱图扫描范围为m/z 200~800,离子传输管温度150℃,喷雾电压3.5kV,透镜电压65.0V,选定离子的隔离宽度为1.6Da,碰撞能量为13%。结果 m/z 756.7的多级质谱图与标准品[DPPC+Na+]完全相符,即肺癌的差异性物质为DPPC。结论 尖端放电喷雾电离质谱技术利用多级质谱分析可以很好地对肺癌和癌旁组织的差异性磷脂进行定性,DPPC可能成为新的肺癌分子标志物,为肺癌的诊断提供新的方法。Objective To identify the differential phospholipid in tung cancer by using tip-spray ionization mass spectrometry (TSI-MS). Methods Under the premise of no pretreatment of lung tissues,the differential phospholipid m/z 756.7 in lung carcinoma tissues and adjacent normal tissues of 14 cases of patients with lung cancer and the standard [DPPC+ Na+] were detected by multistage mass spectrometry using a self-made methanol extraction assisted tip discharge ionization source. The flow rate of metha- nol was 4 uL/min,the ion source was positive ion mode,the the scanning range of single-stage mass spectrometry was m/z 750- 850,the scanning range of multistage mass spectrometry was m/z 200- 800, the ion transport tube temperature was 150 ℃, the spray voltage was 3.5 kV, the lens voltage was 65 V, the selected ion isolation width was 1.6 Da, and the collision energy was 13%. Results The mass spectrum of m/ z 756.7 detected by multistage mass spectrometry was consistent with the standard [DP- PC+Na+ ],that was,DPPC was the differential material of lung cancer. Conclusion TSI-MS can be used to characterize the differential phospholipids in lung cancer tissues and adjacent tissues,DPPC may become a new molecular marker of lung cancer, providing a new method for the diagnosis of lung cancer.
