摘 要:目的研究CLDN9在无功能性垂体腺瘤亚型瘤样细胞瘤中的表达及其与瘤样细胞瘤的发生和侵袭性的相关性。方法收集20例来源于首都医科大学附属北京天坛医院神经外科的瘤样细胞瘤标本(侵袭性和非侵袭性瘤样细胞瘤各10例)和3例正常垂体组织标本。通过实时定量PCR和Westernblot检测CLDN9在正常垂体组织与瘤样细胞瘤之间,以及侵袭性与非侵袭性瘤样细胞瘤之间的表达差异。通过免疫组织化学方法检测Ki-67在瘤样细胞瘤标本中的表达。采用Logistic回归分析检验Ki-67与瘤样细胞瘤侵袭性的相关性。结果实时定量PCR结果显示,在瘤样细胞瘤中CLDN9基因的表达明显高于正常垂体组织(P〈0.01),且在侵袭性瘤样细胞瘤中的表达(6.54±0.32)高于非侵袭性瘤样细胞瘤(2.47±0.12)(P〈0.01);Westernblot检测结果显示,CLDN9蛋白在侵袭性瘤样细胞瘤中的表达明显高于非侵袭性瘤样细胞瘤(3.331±0.165比2.497±0.125,P〈0.01)。Ki-67〉3%的瘤样细胞瘤标本中,侵袭性4例,非侵袭性3例,统计结果显示Ki-57与瘤样细胞瘤的侵袭性无相关胜(P=0.946)。结论CLDN9可能是与瘤样细胞瘤的侵袭性特异相关的一个重要基因。Objective To study the expression of CLDN9 in the oncocytoma subtype of non- functioning pituitary adenomas and the association between CLDN9 and occurrence and invasiveness of oncocytomas. Methods In this study, we selected 20 cases of oncocytomas ( 10 invasive and 10 non- invasive tumors) and 3 cases of normal pituitary glands. The CLDN9 expression levels in all pituitary tumors and normal pituitary glands were detected by quantitative polymerase chain reaction (qPCR) and Western blot. The Ki-67 expression levels were observed by immunohistochemistry. Logistic regression analysis was used to examine the correlation between Ki-67 and the invasiveness of oncocytomas. Results The expression of CLDN9 in oncocytomas was significantly higher than that in normal pituitary tissue (P 〈 0.01 ). CLDN9 mRNA and protein expression levels were higher in invasive oncocytomas compared to those in non-invasive oncocytomas ( P 〈 0.01 ). In the oncocytomas with Ki-67 index over 3%, 4 cases were invasive and 3 cases were non-invasive. The Ki-67 was not correlated to oncocytomas invasiveness (P = 0. 946). Conclusion The gene of CLDN9 may play an important role in invasiveness of oncocytomas as a specific biomarker for invasive oncocytomas.
