摘  要:目的探讨Mirk/Dyrk1B在人配对宫颈癌组织中表达的临床意义及其与其他耐药基因之间的相关性。方法实时荧光定量PCR检测Mirk/Dyrk1B在正常宫颈组织、癌旁组织和宫颈癌组织的表达,免疫组织化学(IHC)检测Mirk/Dyrk1B和3个耐药基因(Pg P、TopoⅡ和GST-π)蛋白在70例宫颈癌组织、40例癌旁宫颈组织和正常宫颈组织的表达水平,分析Dyrkl B和3个编码耐药基因蛋白在配对宫颈癌组织中的表达情况以及临床意义。结果 Mirk/Dyrk1B在宫颈癌组织的表达显著高于癌旁组织和正常宫颈组织。Dyrkl B与组织学分化程度有显著相关性(P<0.05),Dyrk1B与宫颈癌的临床分期和淋巴结转移无显著相关性(P>0.05)。Mirk/Dyrk1B和GST-π存在相关性(P<0.01)。结论Mirk/Dyrk1B在宫颈癌组织中高度表达,并与耐药基因GST-π蛋白表达相关,参与宫颈癌的发生和多药耐药性,可能成为临床早期诊断的肿瘤标记和一个新的目标基因,用于治疗宫颈癌。Objective To explore the expression and clinical significance of Mirk/Dyrk1 B in pairing cervical carcinoma tissues and the correlation among Mirk/Dyrk1 B and other coding proteins of drug-resistant genes. Methods Real-time fluorescent quantitative PCR was employed to detect the expression of Mirk/Dyrkl B in normal cervical tissues,para-carcinoma tissues and cervical carcinoma tissues. Immunohistochemistry( IHC) was employed to detect the expression levels of Mirk/Dyrkl B and 3drug-resistant gene proteins( Pg P,TopoⅡ and GST-π) in 70 cases of cervical carcinoma tissues,40 cases of para-carcinoma tissues and normal cervical tissues,and to analyze Mirk/Dyrkl B and 3 drug-resistant gene associated proteins in matching cervical cancer tissue and clinical significance. Results The expression level of Mirk/Dyrk1 B in expression cervical carcinoma tissues was significantly higher than that in para-carcinoma tissues and normal cervical tissues. There was a significant correlation between the positive rate of Mirk/Dyrkl B and degree of histological differentiation( P 〈 0. 05),but there was no correlation among the positive rate of Mirk/Dyrk1 B and clinical stage and lymph node metastasis of cervical carcinoma( P 〉 0. 05). In addition,the present study showed that Mirk/Dyrk1 B and GST-π had a correlation( P 〈 0. 01). Conclu sion Mirk/Dyrk1 B was highly expressed in cervical carcinoma and had correlation with GST-π,which indicates that it may participate in the occurrence of cervical carcinoma and multi-drug-resistance,and it may become a tumor marker for clinical early diagnosis and a new target gene for treatment of cervical carcinoma tissues.
