摘  要:目的研究南瓜叶不同极性部位的体外抗氧化活性。方法采用系统溶剂法将南瓜叶的乙醇提取物分别用石油醚、氯仿、乙酸乙酯、正丁醇、水等5种不同极性溶剂萃取得到5种萃取物,以抗坏血酸为阳性对照,测定各萃取物的总还原能力、二苯代苦味酰基(DPPH)自由基清除能力、羟自由基清除能力和2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)自由基清除能力,通过比较半清除率浓度(IC_(50))的大小判断各萃取物的抗氧化能力强弱。结果南瓜叶不同极性部位均具有一定的抗氧化性,其中乙酸乙酯极性部位和水提部位的抗氧化能力最强,清除ABTS自由基的IC_(50)分别为0.0389 mg/mL和0.0272 mg/mL,清除羟基自由基的IC_(50)分别为0.2731 mg/mL和0.3474 mg/mL,清除DPPH自由基的IC_(50)分别为0.1847 mg/mL和0.1876 mg/mL,在总还原能力上,乙酸乙酯极性部位和水提部位明显强于其他三个极性部位。同一提取物在不同的评价体系中表现出不同的抗氧化能力,可能与其所含成分及活性氧的性质有关。结论南瓜叶提取物的乙酸乙酯相和水相的抗氧化作用强,是天然抗氧化剂的良好来源。Objective To study the in vitro antioxidant activities of different polarity fractions extracted from Pumpkin leaves. Methods Based on polarity, five extracts were obtained by using different organic solvents including petroleum, chloroform, ethyl acetate, n-butyl alcohol and water. Their antioxidant activities were evaluated by the reduction capa- bility and scavenging capability of DPPH, ABTS, hydroxyl free radical with ascorbic acid. The antioxidant activities of various extracts were compared using half clearance rate (ICs0). Results Each part of extracts from Pumpkin leaves ex- pressed antioxidant activities, while ethyl acetate and water extracts expressed the highest antioxidant activity. The ICs0 for ABTS, "OH and DPPH of ethyl acetate and water extract were 0.0389 mg/mL and 0.0272 mg/mL, 0.2731 mg/mL and 0.3474 mg/mL and 0.1847 mg/mL and 0.1876 mg/mL, respectively. For total reducing capacity, ethyl acetate and water extracts of polar fractions were stronger than other three polar parts. However, the same sample had different an- tioxidant activity with different evaluation method, and it may be related to its components and the nature of reactive oxygen species. Conclusion The ethyl acetate and water fractions of Pumpkin leaves showed strong antioxidant activi- ties and it can be a kind of good source for natural antioxidants.
