摘　　要：目的探讨微小RNA（miRNA，miR）-196a-1在胃癌组织中的表达及其临床意义。方法采用反转录．聚合酶链反应（RT-PCR）检测miR-196a-1在胃癌组织和胃癌细胞株中的表达水平，并分析其与临床病理特征关系。结果RT-PCR结果显示，miR-196a-1在胃癌组织标本的表达量较对照组增加4倍，差异有统计学意义（P〈0．05）；相对于正常胃黏膜上皮细胞株GES-1，胃癌细胞株SGC-7901、AGS、MKN28中的miR-196a-1均高表达，相对表达量分别为：SGC-7901：4．24±0．15（P〈0．05），MKN28：3．17±0．11（P〈0．05），AGS：3．32±0．07（P〈0．05）。胃癌组织miR-196a-1的高表达与患者的肿瘤TNM分期（P〈0．05）、淋巴结转移（P〈0．05）、组织分化程度（P〈0．05）有关，与患者的年龄、性别无关（P〉0．05）。结论miR-196a-1在胃癌组织和胃癌细胞中表达均上调，与胃癌的发生发展明显相关。Objective To discuss the expression of microRNA （ miRNA, miR） - 196a - 1 in gas- tric cancer and its clinical significance. Methods The expression of miR - 196a - 1 in gastric cancer and gastric cancer cell lines was detected by reverse transcriptase -polymerase chain reaction （RT -PCR） and its relationship with clinicopathological features was analyzed. Results The expression of miR - 196a - 1 in gastric cancer tissue was increased by 4 - fold compared with that of the control group （P 〈 0. 05 ） , the difference was statistically significant （P 〈 0. 05 ）; compared with normal gastric epithelial cell line GES - 1, human gastric cancer cell line SGC - 7901, AGS, MKN28 in miR - 196a - 1 were highly ex- pressed. Relative expression levels were： SGC - 7901 ： 4. 24 ± 0. 15 （ P 〈 0. 05 ）, MKN28 ： 3. 17 ± 0. 11 （ P 〈 0. 05 ） , AGS ： 3.32±0. 07 （ P 〈 0. 05 ）. The expression of miR - 196a - 1 in gastric cancer tissue was related to TNM staging （ P 〈 0. 05 ）, lymph node metastasis （ P 〈 0. 05 ） and the degree of histological differentiation （ P 〈 0. 05 ）, which was not related to the age and sex of the patients （ P 〉 0. 05 ）. Conclu- sion The expression of miR - 196a - 1 in gastric cancer and gastric cancer cells was up - regulated, which was significantly related to the occurrence and development of gastric cancer.
