摘  要:目的观察大株红景天注射液对大鼠脑缺血再灌注损伤后血管内皮损伤的影响,并探讨其作用机制。方法 Sprague Dawley大鼠80只,其中24只未造模型大鼠作为对照组;另56只大鼠采用线栓阻塞大脑中动脉法建立脑缺血再灌注损伤模型,术后选择其中造模成功的48只大鼠随机分为模型组和观察组,每组24只。观察组大鼠经尾静脉注射大株红景天注射液2 m L·kg^(-1),每日1次;对照组及模型组大鼠经尾静脉注射等量生理盐水。14 d后断头处死各组大鼠,每组取8只大鼠采用酶联免疫吸附试验检测脑组织血管内皮生长因子(VEGF)、受体胎肝激酶-1(FLK-1)、缺氧诱导因子-1α(HIF-1α)蛋白,另取8只大鼠采用反转录聚合酶链反应测定其脑组织VEGF mRNA、FLK-1mRNA、HIF-1αmRNA表达,余下8只大鼠采用氯化三苯基四氮唑染色法测定脑缺血梗死区比例。结果对照组、模型组、观察组大鼠全脑质量比较差异无统计学意义(P>0.05)。对照组大鼠脑组织无缺血梗死;模型组、观察组大鼠的脑缺血梗死区质量及梗死比例均显著高于对照组(P<0.05);观察组大鼠脑缺血梗死区质量及梗死比例均显著低于模型组(P<0.05)。模型组和观察组大鼠脑组织VEGF、FLK-1、HIF-1α蛋白及其m RNA表达与对照组比较均显著增高(P<0.05);观察组大鼠脑组织VEGF、FLK-1、HIF-1α蛋白及其m RNA表达与模型组比较均显著增高(P<0.05)。结论大株红景天注射液可以升高大鼠缺血脑组织VEGF、FLK-1、HIF-1α蛋白及其m RNA表达,有助于诱导缺血部位血管内皮分化与再生,促进脑缺血区动脉侧支循环重建,维持成熟血管的完整性,增加脑组织供血,对大鼠脑缺血再灌注损伤具有一定的保护作用。Objective To investigate the effect of rhodiola kirilowii Regel injection on rats with vascular endothelial induced by cerebral ischemia reperfusion and to explore the mechanism of action. Methods Among eighty Sprague Dawley rats, 24 rats were not given any operation as the control group, the other 56 rats were performed with occlusion of middle cerebral artery to establish the cerebral ischemia reperfusion models. Forty-eight rats which were built model sucessfully were divided into model group and observation group randomly, with 24 rats in each group. The rats in observation group were given 2 mL · kg^-1 rhodiola kirilowii Regel by caudal intravenous injection, once a day. While the rats in control group and model group were given the same volume of physiological saline. The rats were sacrificed after 14 days. The protein expressions of vascular endothelial growth factor (VEGF), fetal liver receptor kinase 1 ( FLK1 ) and hypoxia inducing factor-1α ( HIF-1α) in brain tissues of 8 rats in each group were detected by enzyme-linked immunosorbent assay. The expressions of VEGF mRNA, FLK-1 mRNA and HIF-1α mRNA of another 8 rats in each group were detected by reverse transcription polymerase chain reaction. Cerebral ischemia infarction area of the left 8 rats was evaluated by triphenyltetrazolium chloride staining method. Results There was no significant difference in total brain mass among the control group, the model group and the observation group ( P 〉 0. 05 ). Ischemia and infarction in brain was not found in the control group. Infarct mass and infarct percentage in brain of model group and observation group were significantly higher than those of control group (P 〈 0. 05). Infarct mass and infarct percentage inbrain of observation group were significantly lower than those of model group ( P 〈 O. 05 ). The protein and mRNA expressions of VEGF,FLK-1 and HIF-lc~ in model group and observation group were significantly higher than those in the control group (P 〈 0.05). The pro
