摘 要:目的探讨表皮生长因子受体(EGFR)抑制剂西妥昔在舌鳞癌多药耐药中的作用。方法 Western blot方法检测舌癌细胞系Tca8113及多药耐药细胞系Tca8113/CBP EGFR的表达,流式细胞术检测EGFR受到抑制后舌癌多药耐药细胞Tca8113/CBP细胞内阿霉素浓度的影响。结果细胞系Tca8113和Tca8113/CBP均有EGFR的表达(P>0.05)。EGFR受到抑制后,Tca8113/CBP细胞内阿霉素浓度升高(P<0.05)。结论 EGFR抑制剂西妥昔可使舌癌多药耐药细胞细胞内药物浓度升高。Objective To investigate the effect of anti-EGFR agents Cetuximab on multidrug resistance (MDR) tongue squamous cell carcinoma cell line TcaS113/CBP. Methods Western blot assay was applied to detect EGFR expression in tongue squamous cell carcinoma cell line TcaS113 and its MDR cell line Tca8113/CBP. The concentration of intracellular drug adriamycin in cell line Tca8113/CBP after application of Cetuximab was measured by flow cytometry. Results EGFR in cell lineTcaSll3 and its MDR cell line TcaSll3/CBP was expressed (P 〉0.05). After the use of anti- EGFR agents Cetuximab, the intracellular drug concentration of adriamycin was significantly increased in MDR cell line Tca8113/CBP (P 〈 0. 05 ). Conclusion The anti--EGFR agent Cetuximab could increase the intracellular drug concentration in MDR cell line TcaSll3/CBP.
