摘 要:目的探讨慢性不可预见刺激(chronically unpredicted stress,CUS)致大鼠抑郁行为与端脑和海马色氨酸羟化酶2(tryptophan hydroxylase-2,TPH2)、多巴脱羧酶(dopa-decarboxylase,DDC)及单胺氧化酶A(monoamine oxidase-A,MAOA)mRNA及蛋白表达的关系。方法 30只♂SD大鼠随机分为模型组(MG)与对照组(CG),每组15只。采用孤养结合CUS连续刺激28 d建立大鼠抑郁模型;以开场实验和强迫游泳实验评价大鼠抑郁行为;采用实时荧光定量PCR与Western blot方法分别检测大鼠端脑和海马TPH2、DDC、MAO-A mRNA及蛋白表达。结果与对照组大鼠相比,模型组大鼠开场实验运动得分明显降低(P<0.01)、强迫游泳不动时间明显延长(P<0.01);端脑和海马TPH2和DDC mRNA及蛋白表达明显下降(P<0.01,P<0.05)、MAO-A mRNA及蛋白表达明显升高(P<0.01,P<0.05)。结论CUS诱导大鼠产生抑郁症样行为,其机制可能与TPH2、DDC及MAO-A异常表达有关。Aim To investigate the role of tryptophan hydroxylase-2( TPH2),dopa-decarboxylase( DDC) andmonoamine oxidase-A( MAO-A) in depression-like behaviors induced by chronic unpredictable stress( CUS). Methods 30 male SD rats were randomly divided into model group( MG) and control group( CG).Rat depression model was developed by CUS for 28 consecutive days in a solitary condition. The depression-like behaviors of rats were evaluated by open-field test( OFT) and forced-swimming test( FST). The real time PCR and Western blot test were used to determine the mRNA and protein expression of TPH2,DDC and MAO-A in rat telencephalon and hippocampus. Results The movement scores of rats were obviously decreased in OFT( P〈0. 01). The immobility time wasobviously increased in FST( P〈0. 01). The mRNA and protein expressions of TPH2 and DDC were decreased significantly( P〈0. 01,P〈0. 05) and the MAO-A mRNA and protein expressions were increased significantly( P〈0. 01,P〈0. 05) in telencephalon and hippocampus of MG rats, when compared with those in CG rats. Conclusion The TPH2,DDC and MAO-A in rat telencephalon and hippocampus were closely related with the depression-like behaviors of rats induced by CUS.
