摘 要:目的观察神经干细胞在体外培养条件下是否有向胶质瘤细胞趋向的特性。方法 1从胎小鼠脑组织中分离出神经干细胞,并采用免疫荧光化学法检测神经干细胞标志物神经上皮干细胞蛋白(Nestin)的表达,免疫细胞化学法检测分化后细胞神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)的表达;2用Transwell细胞小室作神经干细胞对胶质瘤细胞趋向性实验。结果 1从胎鼠脑组织分离的细胞能形成大量悬浮的神经干细胞球,经免疫荧光化学法鉴定,巢蛋白抗体标记阳性,免疫细胞化学法鉴定分化后的神经元、胶质细胞的特异性抗原NSE及GFAP表达阳性;2神经干细胞对胶质瘤细胞趋向实验中,实验组迁移的神经干细胞数明显多于对照组及空白组,三者比较差异有统计学意义(P<0.05)。预诱导神经干细胞对胶质瘤细胞趋向实验中实验组迁移的神经干细胞数明显多于对照组及空白组,差异有统计学意义(P<0.05)。结论该研究观察到神经干细胞在体外有向胶质瘤细胞趋向的特性。预诱导状态下神经干细胞有比未诱导神经干细胞更强的向脑胶质瘤细胞趋向的趋势。Objective To observe whether the neural stem cells have the characteristics of tropism towards glioma cells in vitro. Methods ①Neural stem cells were isolated from the brain tissue of fetal mice. Immunofluorescence was used to detect theexpression of neural stem cell marker-neuroepithelial stem cell protein( Nestin) and the expression of neuron-specific enolase( NSE) as well as glial fibrillary acidic protein( GFAP) after differentiation was detected by immunocytochemical method;②Tropism experiment of neural stem cells towards glioma cells were done with Transwell chambers. Results①Cells isolated from fetal mice brain can form a large number of suspended neural stem cell spheres,which,identified by immunofluorescence,was positively labeled by nestin antibody and identified by immunocytochemical method,positively expressed post-differentiation neuron and gliocyte-specific antigen- NSE and GFAP;②In tropism experiment of neural stem cells towards glioma cells,the number of migrated neural stem cells in the experimental group was significantly more than those in the control group and the blank group,the differences among the three groups being statistically significant( P〈0.05). In tropism experiment of pre-induced neural stem cells towards glioma cells,the number of migrated neural stem cells in the experimental group was significantly more than those in the control group and the blank group,the difference being statistically significant( P〈0.05). Conclusion In this study,the in vitro tropism characteristics of neural stem cells towards glioma cells was observed. Pre-induced neural stem cells have stronger tropism towards brain glioma cells than neural stem cells without pre-induction.
