摘 要:【目的】研究两株SRB菌生长繁殖的影响因素,进行SRB菌剂的初步试制。【方法】采用CFU法进行菌株密度计数,考察不同p H、培养温度、碳源、培养基等条件对两株SRB菌株生长的影响,并对SRB4菌株进行了菌剂的初步试制。【结果】菌株SRB4的最适培养条件:温度为38℃;起始p H为8.0;培养基是以柠檬酸钠为碳源的改良Postgate培养基。菌株SRB3的最适培养条件:温度为36℃;起始p H为7.0;培养基是以玉米芯水提液为碳源的Postgate培养基。经菌剂初步试制,SRB4菌株在发酵48 h后菌株密度保持在1012 CFU/m L以上。【结论】两株SRB菌在最适培养条件下密度保持在1012 CFU/m L以上,可作为工程应用的备选菌株。[Objective] The main factors of the growth of two Sulfate-Reducing Bacterium (SRB) strains were investigated and preliminary trail of SRB microbial inoculum was carried out. [Methods] The impact of pH, temperature, carbon source and culture media on the growth of SRB were studied by the method of CFU for strain density count, moreover, the trial-produce of SRB4 microbial inoculum was implemented. [Results] The optimum culture conditions for SRB4 were as follows: temperature 38 ℃; pH 8.0; provided Postgate medium with sodium citrate as carbon source. The optimmn culture conditions for SRB3 were as follows: temperature 36 ℃;pH 7.0; Postgate medium with the water extract of corn cob as carbon source. The strain density of SRB4 was maintained above 1012 CFU/mL after 48 h femlentation. [Conclusion] Under the optimal conditions,the strain density of the two SRBs were up to 1012 CFU/ mL, and they could be alternative strains for engineering application.
