摘 要:为了对三株未知型牛源产气荚膜梭菌(Clostridium Perfringens,Cl.perfringens)进行基因分型鉴定,将三株未知型菌株进行厌氧培养、纯化,观察培养特性,进行革兰氏染色镜检以及生化管鉴定,根据产气荚膜梭菌α、β、ε和ι毒素基因以及产气荚膜梭菌16S r RNA保守基因分别合成5对引物,建立PCR方法。经培养特性、革兰氏染色镜检、生化特性以及16S r RNA保守基因细菌PCR鉴定均符合产气荚膜梭菌,PCR结果表明,三株产气荚膜梭菌均为A型。Three cattle clostridium perfringens strains which types were unknown were anaerobic cultured,purified and observed culture characteristics, gram stain microscopy and biochemical tube identification were usedto identify their genotypings. PCR method was established based on the clostridium perfringens α,β,ε and ι toxin gene and clostridium perfringens 16SrRNA conserved genes, compounded five pairs primer. Results showed that culture characteristics, gram stain microscopy, biochemical properties, 16S rRNA conservative germs PCR identification accorded with clostridium perfringens .The result of PCR indicated that all three clostridium perfringens strains were type A.
