超声波法和树脂法提取胞外聚合物及其胞外磷的研究

摘  要:以两组实验室规模的厌氧/好氧序批式(A/O-SBR)反应器为研究对象,从胞外聚合物(EPS)聚磷分解、细胞破损率、EPS提取量、胞外磷提取量4个方面,对比研究了超声波法和阳离子交换树脂(CER)法。研究结果表明,超声波法和CER法提取EPS过程均能避免聚磷酸盐分解,同时CER法提取EPS及其胞外磷的效果优于超声波法。超声波法提取EPS及其胞外磷的较优工作条件为超声波功率密度1 W/m L,在空占比为50%的条件下作用6 min,该条件下提取污泥的EPS占污泥有机物(TOCEPS/TOCsludge)的3.62%-9.27%,胞外磷占总磷量(TPEPS/TPsludge)的4.20%-10.57%。CER法提取EPS及其胞外磷的较优工作条件为搅拌速率550 r/min,作用时间30 min,该条件下提取污泥的EPS及其胞外磷分别占污泥有机物(TOCEPS/TOCsludge)与总磷(TPEPS/TPsludge)的10.88%-25.93%和57.09%-61.32%。两组反应器污泥的EPS及其胞外磷的含量存在较大差异。20℃反应器污泥较35℃反应器污泥含有更多的EPS及其胞外磷,且前者的胞外磷中含有更多的聚磷酸盐。With two sets of lab-scale anaerobic/aerobic sequential sequencing reactors(A/O-SBR), extracellular polymers(EPS), phosphorus decomposition, cell breakage, EPS extraction quantity, and extracted extracellular phosphorus were comparatively studied using the sonication method and the cation exchange resin(CER) method. Research results showed that both sonication method and CER method for extracting EPS can avoid polyphosphate decomposition,and the effect of CER on extracting EPS and extracellular phosphorus was better than that of sonication. The optimized conditions of the sonication method for extracting EPS and its extracellular phosphorus were at the ultrasonic power density of 1 w/m L and the idle running ratio of 50% for 6 min. Under such a condition, the extracted EPS accounted for 3.62%-9.27% of organic matter(TOCEPS/TOCsludge) in sludge, and the extracted extracellular phosphorus accounted for 4.20%-10.57% of total phosphorus in sludge(TPEPS/TPsludge). The optimized conditions of the sonication method for extracting EPS and its extracellular phosphorus were that the cation exchange reaction lasted 30 min under 550r/min.Under such a condition, the extracted EPS accounted for 10.88% -25.93% of organic matter in sludge(TOCEPS/TOCsludge), and the extracted extracellular phosphorus accounted for 57.09% -61.32% of total phosphorus in sludge(TPEPS/TPsludge). The sludge EPS and extracellular phosphorus contents between two groups of reactors were largely different. The amounts of sludge EPS and extracellular phosphorus in the reactor at 20℃ were higher than those at35℃. The extracellular phosphorus in the reactor at 20℃ also contained more polyphosphates than that at 35℃.
