摘　　要：目的检测mir-9在新疆维吾尔族和汉族胶质母细胞瘤中的表达,以及探讨异常表达的microRNA与胶质母细胞瘤临床病理学特征的相关性。方法收集25例来源于新疆医科大学第一附属医院2011-2014年手术切除经病理证实的胶质母细胞瘤(WHOⅣ级)患者石蜡组织标本,包括13例汉族和12例维吾尔族,并选取2例癫痫患者切除的正常脑组织作为对照。采用实时定量荧光逆转录聚合酶链反应(RT-PCR)检测mir-9的mRNA水平的表达。用Fisher确切概率法、Cox回归和Graph Pad Prism生存分析方法分析其表达水平与胶质母细胞瘤临床病理特征和预后的关系。结果在胶质母细胞瘤中mir-9有21例显示高表达,占21/25(84.0%)。与对照组相比,mir-9在胶质母细胞瘤中mRNA相对平均表达量显著增高(P<0.05)。不同年龄的患者其mir-9异常表达差异有统计学意义(P<0.05),而不同民族、性别、发病部位的患者mir-9异常表达差异均无统计学意义(P>0.05)。mir-9、化疗、放疗、放化疗联合和年龄与胶质母细胞瘤患者预后相关(P<0.05)。结论 mir-9在胶质母细胞瘤中起着重要的作用,且影响患者的预后,可能作为胶质母细胞瘤的预后指标。Objective To study mir-9 gene mRNA expression in glioblastoma between Uyghur and Han pa-tients in Xinj iang and analyze the correlation of abnormal versions of microRNAs with clinicopathological features of glioblastoma.Methods 25 previously untreated patients （13 Han and 12 Uyghur）who under-went surgical excision of glioblastoma at the First Affiliated Hospital of Xinjiang Medical University from 2011 to 2014 were included,additionally,2 epilepsy patients with normal brain tissue to be control group. The expression of mir-9 was examined by RT-PCR in formalin-fixed paraffin embedded primary tissue specimens.The relationships between the expression of mir-9 and the clinicopathological features of glio-blastoma,and the survival rate of glioblastoma patients were also discussed.The relationship between mir-9 and glioblastoma prognosis was evaluated using a Fisher exact probability test,Cox regression model and Graph Pad Prism survival curve analysis.Results There were 21 cases showed high expression of mir-9, 21/25 （84.0%）.The rates of positive mir-9 RNA expression in glioblastoma were significant higher than＆amp;nbsp;those in control （P 〈0.05）.The expression of mir-9 was correlated with patient age （P 〈0.05）while not correlated with patient nationality,gender,location （P 〉0.05）.Single factor analysis determined that high expression of mir-9,chemotherapy,radiation therapy,radiochemotherapy and patient onset age were associated with glioblastoma patient prognosis （P 〈0.05）.Conclusion The expression of mir-9 plays a role in glioblastoma progression,and may be used as a prognostic marker in glioblastoma.
