猪圆环病毒2型河南株全基因组的克隆与遗传进化分析

摘　　要：为了解近几年河南地区猪圆环病毒2型（PCV2）的遗传进化情况,本研究对采自2011年-2015年河南省13个地区的28份疑似患断奶仔猪多系统衰竭综合征（PMWS）猪的病料样品进行PCR检测,并将检出的阳性病料样品接种无PCV的PK-15细胞,分离PCV2,进而用IPMA进行鉴定。最后对所获分离株进行全基因组克隆和序列分析。结果显示,28份样品中阳性率为75%（21/28）,并成功分离出21株PCV2,其全基因序列同源性为95.4%-99.9%;在遗传演化关系上,21株PCV2分离株分布于两个基因型（PCV2b和PCV2d）,16株属于PCV2d,而其余5株属于PCV2b基因型,这也说明PCV2d逐渐成为河南地区的优势流行株。此外,通过对其Cap蛋白氨基酸序列的预测与分析,发现其主要抗原区存在12处位点明显变异,这可能影响其抗原性及免疫原性。本研究为河南地区PCV2的防控提供了依据。To investigate the genetic variation of porcine circovirus type 2 （PCV2） in recent years in Henan, twenty-eight samples suspiciously infected with PCV2 were collected from 13 regions from 2011 to 2015, and subjected to detection by PCR and virus isolation in PK-15 cells. The results showed that the PCV2 positive rate was 75% （21/28）, and the 21 isolates of PCV2 were isolated with PK-15 cells and sequenced. The homology was 95.4% to 99.9% among 21 isolates. The phylogenetic analysis showed the 21 PCV2 were classified into two genotypes （5 PCV2b and 16 PCV2d）, indicating that PCV2d was gradually become the predominant epidemic strains in Henan. Moreover, 12 major heterogenic regions were found in the main antigen region of capsid protein, which might affect the antigenic and the immunogenicity of PCV2. These results provided the basis for the prevention and control of PCV2 infection in Henan.
