摘  要:目的建立高效液相色谱法测定跌打活血酊中羟基红花黄色素A和阿魏酸的含量。方法使用ODS2 C_(18)(4.6 mm×150 mm,5μm)色谱柱,其中羟基红花黄色素A的色谱条件:以甲醇-0.7%磷酸溶液(25∶75)为流动相,流速为1 m L·min^(-1),检测波长为403 nm,柱温为30℃,进样量为10μL;阿魏酸的色谱条件:以乙腈-0.085%磷酸溶液(17∶83)为流动相,流速为1 m L·min^(-1),检测波长为316 nm,柱温为35℃,进样量为10μL。结果羟基红花黄色素A在0.0478~2.39μg与峰面积线性关系良好(r^2=0.9996),样品的平均加样回收率为98.6%;阿魏酸在0.028 35~2.835μg与峰面积线性关系良好(r^2=0.9998),样品的平均加样回收率为99.1%。结论建立的方法具有操作简便、结果准确、灵敏度高等优点,可用于控制跌打活血酊的质量。Objective To establish an HPLC method to determine the content of hydroxy safflower yellow A and ferulic acid in Dieda Huoxue tincture. Methods ODS2 C18(4.6 mm×150 mm, 5 μm) was used as chromatographic column. Chromatographic conditions for hydroxy safflower yellow A: methanol-0.7% phosphoric acid solution(25∶75) as the mobile phase, with the flow rate 1 m L·min^-1, detection wavelength 403 nm, column temperature 30 ℃, and injection volume 10 μL. Chromatographic conditions for ferulic acid: acetonitrile-0.085% phosphoric acid solution(17∶83) as the mobile phase, with the flow rate 1 m L·min^-1, detection wavelength 316 nm, column temperature 35 ℃, and injection volume 10 μL. Results Hydroxy safflower yellow A had a good linearity at 0.0478- 2.39 μg(r^2= 0.9996), and the average recovery was 98.6%. Ferulic acid had a good linearity at 0.028 35- 2.835 μg(r^2= 0.9998), and the average recovery was 99.1%. Conclusion The method is simple, accurate, and highly sensitive, and can be used for the quality control of Dieda Huoxue tincture.
