摘  要:目的:通过观察苏木乙酸乙酯提取物对急性病毒性心肌炎小鼠模型心肌组织中干扰素-γ(Interferon gamma,IFN-γ)水平及其心肌组织病理形态学改变的影响,探讨苏木对病毒性心肌炎的治疗机制。方法:选用清洁级、4周龄近交系纯种健康雄性Balb/c小鼠50只,随机分为3组,苏木乙酸乙酯提取物组20只,模型对照组20只,空白对照组10只,苏木乙酸乙酯提取物组、模型对照组小鼠给予腹腔注射接种半数组织培养感染剂量为10-7/m L的柯萨奇病毒B3(CVB3)Nancy株病毒稀释液0.3 m L。空白对照组小鼠注射同体积的RM1640溶液;接种病毒0.5 h后,苏木乙酸乙酯提取物组小鼠给药苏木乙酸乙酯提取物溶液0.2 m L;模型对照组及空白对照组按体质量计算等容量0.2%羧甲基纤维素钠溶液给药,每日1次,连续给药14 d后,小鼠断颈处死并取出心脏,常规石蜡包埋,采用免疫组化法测定小鼠心肌组织IFN-γ的含量,光镜下观察心肌病理形态学改变。结果:与模型对照组比较,苏木乙酸乙酯提取物组小鼠心肌组织IFN-γ的表达水平明显降低(P<0.05)。结论:苏木乙酸乙酯提取物能够显著降低CVB3感染小鼠心肌组织中IFN-γ表达水平,从而发挥治疗病毒性心肌炎的作用。Objective: To observe the ethyl acetate extracts from hematoxylin acute viral myocarditis in mice models according to myocardial tissue levels of IFN - γ and myocardial pathological changes, and to explore hematoxylin for therapeutic mechanism of viral myocarditis. Methods : Clean grade, four - week old purebred male inbred Balb/c mice were randomly divided into three groups, hematoxylin ethyl acetate extract group with 20 mice, model control group with 20 mice, control group with 10 mice. In hematoxylin ethyl acetate extract group and model control group mice with abdominal iodophor disinfection were injected with intraperitoneally inoculated tissue culture infective dose half of 107/mL of coxsackievirus B3 ( CVB3 ) Nancy strain virus dilution 0.3 mL. In blank control group the mice were injected with the same volume of RM1640 solution; 0.5 hours after inoculation, the ethyl acetate extract hematoxylin mice were fed with the ethyl acetate extract solution 0.2 mL; in model control group and blank control group according to body weight the computing capacity was of 0.2% sodium carboxymethyl cellulose solution gavage ; once daily orally for 14 days, and the mice were sacrificed and mice's hearts were removed, heart specimens conventional embedded in paraffin, determined by immunohistochemistry cardiac muscle tissue IFN -γ levels under light microscope myocardial pathological changes. Results : Compared with the model group, the expressions of IFN - γ levels were significantly lower in mice of hematoxylin ethy acetate extract group ( P 〈 0.05 ). Conclusion : Hematoxylin ethyl acetate extract can reduce the viral infection myocardial pathological damage of myocardial tissue, which has a protective effect.
