摘 要:以乙二醇预处理的棉花秸秆为试验材料,以酶解时间、酶解温度、pH、底物质量浓度和纤维素酶浓度为试验因素,设计单因素试验、Box-Behnken试验,考察各因素对还原糖质量分数的影响,优化酶解糖化条件。结果表明,纤维素酶浓度和酶解时间对还原糖质量分数的影响极显著,二者的交互作用对还原糖质量分数有显著影响。还原糖质量分数与纤维素酶浓度、底物质量浓度、酶解时间之间的回归模型有统计学意义,各因素的影响主次顺序为纤维素酶浓度>酶解时间>底物质量浓度。酶解糖化优化条件为纤维素酶浓度90FPU/g、底物质量浓度47.2g/L、50℃、pH 4.8下糖化72h,还原糖质量分数高达486.9mg/g,显著高于原秸秆糖化效果(178.2mg/g)。The enzymatic saccharification parameters of cotton stalk pretreated by ethylene glycol were investigated.The effect of hydrolysis time,hydrolysis temperature,pH,substrate mass concentration,and cellulase concentration on reducing sugar mass fraction was studied by the single factor experiment and Box-Behnken design to optimize enzymatic saccharification conditions.The results indicated that the influence of cellulase concentration and hydrolysis time on reducing sugar mass fraction was highly significant,and the interaction between the two factors had a significant influence on the reducing sugars mass fraction.The model obtained had a statistically significance.The influences on reducing sugars mass fraction followed the order of cellulase concentrationhydrolysis timesubstrate concentration.The reducing sugars mass fraction of pretreated cotton stalk was 486.9mg/g,significantly higher than the native biomass(178.2mg/g)under the optimized conditions of cellulase concentration 90FPU/g,substrate concentration 47.2g/L,hydrolysis time 72 h,hydrolysis temperature50 ℃ and pH 4.8.
