Twist介导上皮间质转化参与胃癌干样细胞的形成及其相关机制的研究

摘　　要：目的研究Twist蛋白在胃癌干样细胞向上皮间质转化过程中的作用机制。方法采用免疫组化检测方法对42例胃癌标本中的Twist蛋白、人低氧诱导因子1α(HIF-1α)水平进行检测;并对同期体检健康人群胃黏膜组织中的Twist蛋白、HIF-1α水平进行检测。比较两组研究对象的Twist蛋白、HIF-1α阳性率,分析其与胃癌干样细胞转化上皮间质的关系。结果胃癌组Twist蛋白阳性率(59.52%)明显高于对照组(16.67%),胃癌组HIF-1α阳性率(64.29%)亦明显高于对照组(21.43%),差异均有显著性(P<0.05)。胃癌患者Twist、HIF-1α的表达与淋巴转移、分化程度、浸润深度、淋巴结转移和远处转移有密切关系(P<0.05),但与年龄、性别和肿瘤大小无明显相关性(P>0.05)。结论 Twist蛋白的表达参与胃癌干样细胞向上皮间质转化的过程,其表达与胃癌的转移和侵犯相关,对胃癌的诊断、治疗和预后具有一定的指导意义。Objective To study the mechanism of Twist protein involved in the mechanism and process of gastric stem cells to epithelial-mesenchymal transformation.Methods The levels of Twist protein and HIF- 1α in specimens of 42 patients with gastric cancer during May 2014 to May 2016 were examined by using immunohistochemical method,and compared with those of gastric mucosa in physically healthy persons,HIF-1α levels were detected.The positive rates of Twist protein and HIF- 1α were analyzed,and the relationship between these markers with dry samples of gastric epithelial- mesenchymal transformation had been studied.Results The difference in positive rates of Twist and HIF- 1α between patients with gastric cancer and healthy persons was statistically significant（P 〈0.05）;and the expression levels of Twist protein and HIF- 1α in patients with gastric cancer were closely correlated with the metastasis in lymph nodes and the expression of HIF- 1α in patients with gastric cancer and the degree of differentiation of gastric cancer and distant metastasis（P 〈0.05）,but there was no signficant correlation with age,gender and tumor size（P〉 0.05）.Conclusion The expression of Twist protein is participated in the process of gastric cancer stem cells to epithelial- mesenchymal transformation,its expression is correlated with the metastasis and violation of gastric cancer,hence it has certain significance in diagnosis,treatment and prognosis of gastric cancer.
