摘  要:通过PCR检测食用油中内源性DNA是鉴定食用植物油品种真实属性,检测食用油掺假的一种有效方法。然而由于精炼的食用油中DNA含量极低,如何有效富集食用油中的DNA是实现PCR鉴别粮油的关键。因此,作者研究了磁珠法富集食用油中的DNA并通过实时荧光PCR检测芝麻油、菜籽油、稻米油的内源基因。结果表明经过5次乳化后和磁珠进一步富集水相中的DNA,实时荧光PCR成功检测到了食用油中的内源性基因,琼脂糖凝胶电泳分析进一步证明实现了上述3种食用植物油的定性鉴定。The detection of endogenous genes using PCR is one of the effective methods to detect the variety of edible vegetable oil and identify their adulteration. Due to the extremely low content of DNA in the refined oil, the effective extraction method to concentrate DNA is the crucial challenge concerning the authenticity identification based on PCR. The DNA concentrated method based on magnetic beads was established in this paper. DNA was extracted using this method from the edible oil including sesame oil, rapeseed oil and rice oil. Their endogenous genes was then characterized by real-time fluorescence PCR.The endogenous genes were successfully detected using concentrated DNA extracted after five emulsifications and further by magnetic beads. The results were further confirmed by the agarose gel electrophoresis. The qualitative identification of edible oil using magnetic beads was achieved.
