摘 要:通过对蛹虫草基因组数据挖掘,发现Jacalin类凝集素编码基因JRL-ccm4,进而对该基因及其编码产物进行了生物信息学分析,通过原核异源表达获得凝集素蛋白JRL-ccm4,并检测了表达产物的生物学活性测定;通过RT-PCR对不同生长时期的蛹虫草样品中JRL-ccm4基因的表达水平进行了检测。研究结果表明:JRL-ccm4蛋白有一个JRL结构域,能在原核细胞中成功表达;异源表达的JRL-ccm4蛋白具有抑制金黄色葡萄球菌的活性,有一定抑制人宫颈癌细胞HeLa增殖的活性;原基期JRL-ccm4基因表达水平最高,是菌丝期的3.49倍,是子实体时期的2.39倍,表明JRL-ccm4基因可能参与蛹虫草子实体形成过程。Through the Cordyceps militaris genome data mining, a lectin gene encoding JRL-ccm4 was found. In this research, the gene and its encoding product were analyzed by bioinformatics, and heterologously expressed and examined the biological activity of the protein in Escherichia coll. The JRL-ccm4 gene expression level in different growth periods of C. militaris was tested by qRT-PCR. The results showed that the JRL-ccm4 proteins had a JRL domain structure which could be successfully expressed in prokaryotic ceils. The protein of the JRL-ccm4 could inhibit growth of Staphylococcus aureus and partly inhibit cervical cancer HeLa cell proliferation ; the JRL-ccm4 had the highest amount of gene expression in stromata, which was 3.49 times that of hyphae period and 2.39 times in the period of the fruiting body. It suggested that the gene could be related to fruit body development.
