摘  要:目的:研究通过肺腺癌 A549细胞构建的裸鼠肺癌组织中 S100A2的表达。方法将30只裸鼠随机平均分为对照组及处理组,处理组采用气管内注入 A549细胞的方法构建裸鼠肺癌动物模型,对照组换用等量氯化钠注射液。通过实时荧光定量 PCR 法和 Western - blot 法检测肺癌组织中 S100A2 mRNA 和 S100A2蛋白的相对表达量。结果对照组裸鼠死亡1例,处理组小鼠死亡3只。处理组裸鼠成功构建肺癌模型。处理组肺癌组织中S100A2 mRNA 相对表达量是对照组的(2.126±0.076)倍,差异有统计学意义(P <0.05)。处理组肺癌组织中S100A2蛋白的相对表达量较对照组升高,差异有统计学意义(P <0.05)。结论裸鼠肺癌组织中 S100A2的表达增高,为 S100A2成为肺癌患者筛查的标志物提供了实验室依据。Objective To study the expression of S100A2 of lung cancer xenografts in nude mice. Methods Thirty nude mice were randomly divided into control group and treatment group. The lung cancer model was established by intratracheal injection of A549 cells in the treatment group,and the control group was treated with the same volume of normal saline. The expressions of S100A2 mRNA and S100A2 protein were measured by real - time PCR method and western - blot method separately. Results One mouse died in the control group and three mice died in the treatment group. The lung cancer model was successfully established in nude mice. The relative expression of S100A2 mRNA in the lung cancer tissues was(2. 126 ± 0. 076)times that of the control group,the difference was significant (P 〈 0. 05). The relative expression of S100A2 protein in lung cancer tissues was significantly higher than that in the control group(P 〈 0. 05). Conclusions The expression of S100A2 in lung cancer tis-sues of nude mice is increased,which provided the laboratory evidence for S100A2 as a marker of lung cancer screening.
