摘  要:为了探讨肿瘤相关巨噬细胞(tumor associated macrophages,TAMs)在人子宫颈癌微环境内的分布与T细胞亚型和临床病理特征的相关性,本研究应用S-P免疫组化法检测TAMs在60例子宫颈癌组织、10例高级别鳞状上皮内病变组织及10例子宫颈组织的分布情况;间接免疫荧光双标法分析浸润的FOXP3^+Treg细胞数量、CD8^+T细胞数量及其粒酶B分泌情况。统计学分析TAMs数量与癌间质浸润的FOXP3^+Treg、CD8^+T数量和临床病理特征的相关性。结果显示TAMs在子宫颈癌微环境的数量均显著多于高级别鳞状上皮内病变和子宫颈组织(p<0.01,p<0.01),同时高级别鳞状上皮内病变组织阳性细胞数也明显多于子宫颈组织(p<0.01);TAMs数量与宫颈癌间质内浸润的FOXP3^+T细胞数量呈正相关性(p<0.05),且与淋巴结转移和癌栓相关(p<0.05,p<0.05),但与CD8^+T细胞浸润数量无明显相关性(p>0.05)。本实验发现TAMs在宫颈癌微环境内大量分布,且与肿瘤局部免疫抑制及淋巴结转移和癌栓有关。The objectives of this research were to investigate the distribution of tumor associated macrophages (TAMs) and its correlation with the number of infiltrating T lymphocytes as well as clinicopathological features in human cervical cancer. The distribution of TAMs was detected in 60 cases of cervical cancer, 10 cases of high- grade squamous intraepithelial lesion (HSIL) and 10 cases of cervical tissue using immunohistochemical staning. Tumor-infiltrating FOXP3^+T, CD8^+T lymphocytes and its levels of Granzyme B production were determined through immunofluorescent double-staining. Correlation between the number of TAMs and that of tumor- infiltrating FOXP3^+T, CD8^+T as well as clinicopathological features was analyzed. Immunohistochemical and immunofluorescent staining showed that TAMs were distributed densely in cervical cancer, the number of them was significantly higher than that in HSIL and cervical tissue (p 〈0.01, p 〈0.01), and positively correlated with that ofFOXP3^+Treg (p〈0.05) as well as lymphatic metastasis and cancer embolus (p〈0.05, p〈0.05), but had no relationship with CD8^+T. We found that TAMs were distributed densely and associated with local antitumor immune inhibition in cervical cancer, as well as lymphatic metastasis and cancer embolus.
