潮湿环境下大鼠脾组织差异蛋白组学研究

摘　　要：目的:通过比较正常及潮湿环境下大鼠脾组织蛋白质谱,筛选和鉴定与湿邪有关的疾病特异性蛋白,探寻湿邪致病的生物标志物。方法:选取SD大鼠20只随机分为对照组、外湿组,每组10只。造模成功后无菌摘除脾脏,取出脾组织10mg/只,进行双向电泳实验、双向凝胶电泳图象分析,肽质量指纹图谱与电喷雾串联质谱鉴定蛋白质,分析差异蛋白质意义。结果:通过比较正常及潮湿环境下大鼠脾组织差异蛋白质,发现3个有意义的蛋白质:表达上调的有2个:载脂蛋白A-I、肌动蛋白相关蛋白2/3复合物亚基5;表达下调的有1个:结合珠蛋白。结论:正常及潮湿环境下大鼠脾组织蛋白质存在差异表达,这些差异蛋白质可能是与湿邪致病相关的疾病特异性蛋白。Objective：To screen and identify the pathogenic specific proteins associated with dampness by comparing the protein spectra of spleen tissue of rats in normal environment and that in humid environment.Methods：Twenty SD rats were randomly divided into the control group and the external dampness group,with 10 rats in each group.After successfully modeling and aseptic extraction of the rat spleen,10 mg spleen tissue of each was extracted.Protein was identified through twodimensional electrophoresis,two-dimensional gel electrophoresis image analysis,peptide mass fingerprinting and electrospray ionization tandem mass spectrometry（ESI MS/MS）,and the significance of differential protein was analyzed.Results：Through the comparison between rat spleen tissue protein in normal environment and that in humid environment,three kinds of pathogenic protein were discovered.Two of them were upregulated in the expressions,which were apolipoprotein A-I and actin associated protein 2/3 compound subunit 5,while one expression was down regulated,identified as haptoglobin.Conclusion：There exist differences between the spleen protein expression of rats in normal environment and that in humid environment,and these differential proteins possibly are pathogenic specific proteins related to the dampness.
