摘 要:目的探讨Toll样受体(Toll-like receptor,TLR)2与TLR4在溃疡性结肠炎(ulcerative colitis,UC)SD大鼠结肠黏膜上皮细胞的表达情况,及维生素D3对UC的治疗作用。方法 SD大鼠60只,随机分为正常对照组、UC模型组和维生素D3治疗组,每组20只。UC模型组、维生素D3治疗组采用三硝基苯磺酸法连续灌肠10d,建立UC模型。造模完成后,维生素D3治疗组采用胆维丁乳原液灌胃,按0.005mL/g,1次/d,连用9d;正常对照组和UC模型组大鼠给予生理盐水灌胃9d。各组大鼠均行疾病活动指数(disease activity index,DAI)评分,并对大鼠结肠组织行组织病理学评分(histopathology score,HPS),采用实时荧光定量PCR法检测结肠组织TLR2 mRNA与TLR4 mRNA表达情况。结果 UC模型组DAI评分(8.06±1.95)、HPS评分(14.56±0.84)和结肠组织TLR2mRNA(0.420 00±0.170 00)、TLR4mRNA(1.580 00±1.000 00)表达水平均明显高于正常对照组(0、0、0.000 38±0.000 18、0.000 57±0.000 23)和维生素D3治疗组(5.09±2.16、10.12±2.09、0.027 00±0.019 00、0.018 00±0.015 00)(P<0.05),维生素D3治疗组高于正常对照组(P<0.05)。结论维生素D3能部分抑制TLR2与TLR4表达,对UC有辅助治疗作用。Objective To explore the expression of Toll-like receptor(TLR)2and TLR4 of intestinal epithelial cell in rat models with ulcerative colitis(UC)and the effect of vitamin D3(VitD3)on UC.Methods Sixty SD rats were randomly divided into control group,model group and VitD3 group,with 20 rats in each group.Model group and VitD3 group were given trinitrobenzene sulfonic acid for 10 dto establish UC models.And VitD3 group received oral gavage of VitD30.005mL/g once a day,totally for 9d.Control group and model group received gavage of normal saline for 9 d.The histopathology score(HPS)and disease activity index(DAI)score were evaluated and the expressions of TLR2 mRNA and TLR4 mRNA were detected by real-time fluorescence quantitative PCR.Results DAI score(8.06±1.95),HPS score(14.56±0.84)and the expressions of TLR2 mRNA and TLR4 mRNA(0.420 00±0.170 00,1.580 00±1.00000)in model group were significantly higher than those in control group(0,0,0.000 38±0.000 18,0.000 57±0.00023)and VitD3 group(5.09±2.16,10.12±2.09,0.027 00±0.019 00,0.018 00±0.015 00)(P〈0.05),and were significantly higher in VitD3 group than those in control group(P〈0.05).Conclusion VitD3 can partially inhibit the expression of TLR2 and TLR4,and may contribute to the treatment of UC.
