摘 要:目的:探讨不同异种脱细胞真皮基质对内皮细胞血管生成的促进作用。方法:利用人脐静脉内皮细胞(HUVEC),与猪脱细胞真皮基质(P-ADM)和牛脱细胞真皮基质(B-ADM)共培养,以Bio-Gide可吸收生物膜和空白组作对照,ELISA法分别检测细胞接种4、6、12、24、48、96 h共培物上清中血管内皮生长因子(VEGF)的含量;GFP慢病毒转染于HUVEC后,在倒置相差显微镜下计数共培物上细胞管型的数目。结果:在细胞接种24 h时,ADM组与Bio-Gide组和空白组比较VEGF的浓度更高(P〈0.05);48 h时P-ADM组VEGF的浓度高于与Bio-Gide组和空白组(P〈0.05)。转染GFP慢病毒的HUVEC与生物膜共培养6~48 h时,B-ADM组细胞管型多于P-ADM组和Bio-Gide组;24 h内,P-ADM组多于Bio-Gide组;空白组未见明显管型。结论:P-ADM与B-ADM比Bio-Gide具有更好的成血管作用,且B-ADM效果更明显,提示异种ADM有较好的促新生血管的作用,可作为替代物应用于膜龈手术。Objective:To research the effect of xeno-acellular dermal matrix(xeno-ADM) on vasculogenesis of human umbilical vein endothelial cell(HUVEC).Methods:Porcine acellular dermal matrix(P-ADM) and bovine acellular dermal matrix(B-ADM)were co-cultured with HUVEC,using blank and Medical collagen membrane(Bio-Gide) as control.The concentration of vascular endothelial growth factor(VEGF) collected from different groups at 4,6,12,24,48,96 h was determined by ELISA test.Meanwhile,the number of cellular casts were counted by observing HUVEC infected by GFP expression lentivirus.Results:The VEGF concentration of xeno-ADM groups were higher than that of the control groups(P〈0.05) after 24 h,and P-ADM was more than that of the control groups after 48 h(P〈0.05).The number of cellular casts of P-ADM group was about as twice as that of the control group after 24 h.The B-ADM group improved less than the P-ADM group.No cellular cast was found in the control group.Conclusions:Xeno-ADM has better effect on vasculogenesis than traditional medical collagen membrane,which suggesting that it has the potential to be the next generation of surgical material applying to mucogingival therapy.
