摘 要:罗伊氏乳杆菌(Lactobacillus reuteri)常栖息于人和动物的肠道系统中,能产生一种特殊的抑菌物质——罗伊氏素,是调节肠道功能的益生菌.乳酸菌活菌制剂广泛应用于食品、医疗、饲料等相关行业.喷雾干燥的能耗较低,在同样生产出优质产品的前提下,冷冻干燥消耗的能量是喷雾干燥的6~10倍.本文通过吸附材料的筛选、保护剂的选择,并对喷雾干燥相关条件进行优化,利用优化的条件进行罗伊氏乳杆菌制剂的制备,并对制备的乳酸菌制剂进行热稳定性实验以及相关质量测定(电镜观察、水分含量、粒径、得率等).结果表明,利用改性硅藻土作为吸附材料的乳酸菌制剂活菌存活率优于改性沸石,选定脱脂牛奶和果聚糖作为保护剂,最佳喷雾干燥条件为:进口温度120?C,出口温度60?C,脱脂牛奶10 g,果聚糖5 g,硅藻土5 g,通过此优化条件制备的乳酸菌制剂其活菌存活率达到88%,活菌数达到5.14×109CFU/g.该乳酸菌制剂在70?C条件下水浴1h,乳酸菌活菌数为3.49×106CFU/g.研究表明,通过优化的喷雾干燥条件制备的乳酸菌制剂存活率高,热稳定性较好.Lactobacillus reuteri often exists in the intestinal system of human and animals, which can produce a special kind of antibacterial substances-reuterin. Lactic acid bacteria living bacteria preparation is widely used in food, health care, feed and other related industries. Spray drying is of low energy consumption,the consumption of freeze drying energy is 6-10 times of spray drying under the same condition of good products.This paper selecting adsorption materials and protective agent, optimizing related conditions of spray drying, and then using the optimized conditions for preparation of lactobacillus reuteri, finally measuring the quality and do the thermal stability experiments of agent of lactobacillus reuteri(electron microscope,moisture content, particle size, yield, etc.). The research show that the survival rate is better when use the modified diatomite as adsorption material of lactic acid bacteria preparation than modified zeolite. When choose skim milk and fructan as protectant, the best spray drying conditions are:inlet temperature 120℃,outlet temperature 60℃, skim milk, 10 g, fructan 5g,diatomite 5g. Through the optimization conditions of preparation of lactobacillus preparation, the living bacterium survival rate will reach 88%, and the number of living bacterium will achieve 5.14 x 10^9CFU/g. When change the lactic acid bacteria preparation under the condition of 70℃ water bath for 1 h, lactic acid bacteria living bacterium number will be 3.49 x 10^6CFU/g.Therefore, the study finally conclude that through the optimization of the spray drying conditions will not only improve the survival rate of lactic acid bacteria, but also enhance the thermal stability.
