摘 要:〔目的〕探索热疗对原代细胞系及耐药骨肉瘤细胞系成瘤体积的影响,并用免疫组化方法检测不同细胞系热疗后HSP 70基因表达。〔方法〕原代细胞系和耐药细胞系接种小鼠16只,按体积相似分成4组。实验组用热疗43℃30 min对照组未进行热疗处理,根据肿瘤体积大小及生存时间绘制曲线。用免疫组化方法测定热疗后不同细胞系成瘤的组织在HSP 70的基因表达情况。〔结果〕热疗处理后肿瘤生长与对照组相比有明显的差异(P<0.05)。耐药组与原代组比较未见明显差异(P>0.05);热疗后实验组肿瘤组织HSP 70蛋白表达明显高于未热疗处理组实验小鼠组织(P<0.05),Saos-2细胞系成瘤鼠组和Saos-2/MTX4.4细胞系成瘤鼠组热处理和无热处理组间未见明显差异(P>0.05)。〔结论〕热疗对耐药细胞系和原代细胞系成瘤均有杀伤作用。热疗后两种细胞系都表现出HSP 70表达升高。〔O bjective〕The antiosteosarcoma effects of hyperthermia were explored by detecting tumor cell volume and HSP 70 gene expression in different cell lines.〔M ethods〕Sixteen mice were inoculated into the primary cell line and drug resistant cell line. These mice were divided into 4 groups according to their volume. The hyperthermia treatment group was treated at 43 ℃ for30 min. The expression of HSP 70 gene in different tumor cell lines was determined using immunohistochemistry. 〔R esults〕Compared with the control group, the tumor growth was significantly different after hyperthermia treatment( P〈0. 05) in the experimental group. The expression of HSP 70 protein in the experimental group was significantly higher than that in the untreated group( P〈0. 05), but the difference was not significant( P〉0. 05)( P〈0. 05), and there was no significant difference between heat treated group and non-heat-treated group( P〈0. 05).〔C onclusion 〕Hyperthermia has the killing effect on the tumor cell lines and the primary cell lines. The hyperthermia treatment can increase the HSP 70 expression i in the same cell line.
