摘 要:目的探讨全血标本离心前不同温度与时间对人巨细胞病毒(human cytomegalovirus,HCMV)核酸检测(nucleic acid amplification test,NAT)结果的影响,以确定离心前放置的安全时限。方法采集含高、中、低DNA载量的全血。采血后按保存温度分为4℃、25℃、37℃组,各组放置0、1、6、12、24、48、72 h后进行离心,行实时荧光定量PCR。结果高浓度4℃组离心前放置72 h,25℃与37℃组离心前放置24 h,无明显变化;中浓度4℃组离心前放置24 h,25℃组、37℃组离心前放置12 h,无明显变化;低浓度4℃组、25℃组离心前放置12 h,37℃组离心前放置6 h,无明显变化。结论为保证HCMV NAT结果的准确性,全血标本最好在离心前4℃保存,并在采血后6 h内及时离心送检,高温条件下更应注意。Objective To investigate the effect of storage temperature and time of whole blood samples before centrifugation on the HCMV NAT results. Methods Once the whole blood samples were collected,they were stored at 4 ℃,25 ℃and 37 ℃,for 0,1,6,12,24,48 h and 72 h before their centrifugation,and were tested by real time fluorescence quantitative PCR. Results In the high copies group which was stored at 4 ℃ from 0 h to 72 h,and stored at 25 ℃ and 37 ℃,respectively,from 0 h to 24 h,there was no significant change. In the medium copies group which was stored at 4℃ from 0 h to 24 h,and stored at 25 ℃ and 37 ℃,respectively,from 0 h to 12 h,there was no significant change. In the low copies group which was stored at 4 ℃ and 25 ℃,respectively,from 0 h to 12 h,at37 ℃ from 0 h to 6 h,there was no significant change. Conclusion In order to ensure the accuracy of HCMV NAT,the samples should best be stored at 4 ℃ and centrifuged within 6 hours after blood collection,especially under the condition of high temperature.
