摘 要:目的为了提高拉沙里链霉菌Streptomyces lasaliensis产拉沙里菌素的发酵水平,利用响应面法对其发酵条件进行优化。方法用Plackett-Burman实验对影响发酵水平的9个主要因素进行显著性评价,筛选出3个显著因素,再选取最陡爬坡法逼近最佳响应面区域的发酵条件,通过CCD中心组合实验以及响应面分析,确定最优发酵条件。结果优化后,最佳培养条件为葡萄糖6.26g/L、糊精28.75g/L、p H 5.58,最佳产量为393.121mg/L。结论在此优化条件下,拉沙里菌素的产量提高了1.18倍。Objective To improve the fermentation level of Streptomyces lasaliensis production of Lasalocid, the response surface method was applied to optimize the fermentation conditions. Methods The significance of the nine main factors affecting the fermentation level was evaluated by Plackett-Burman test, three significant factors were selected, and then the fermentation conditions of the optimal response surface area were approached by the steepest ascent method. Through central composite experiments and response surface analysis, the optimum fermentation conditions were determined. Results After optimization, the optimal culture conditions were glucose 6.26g/L, dextrin 28.75g/L, pH of 5.58, and the optimum yield was 393.121mg/L. Conclusion Under optimal conditions, lasalocid production increased 1. 18 times.
