Determination of Volatile Oil Components in Wild and Cultivated Ningguo Peucedanum praeruptorum Dunn by Gas Chromatography-Mass Spectrometry

摘  要:目的采用气相色谱-质谱联用法(gas chromatography-mass spectrometry,GC-MS)分析不同生长条件下宁前胡中挥发油成分,研究野生和栽培宁前胡挥发油成分含量及种类的区别。方法以安徽省宁国市中溪镇野生和栽培宁前胡、宁国市万家乡镇栽培宁前胡、宁国市栽培宁前胡(统货)为样品,采用《中华人民共和国药典》一部甲法提取挥发油,GC-MS分析挥发油化学成分,归一化法计算各组分的相对含量。结果宁国市中溪镇野生宁前胡总挥发油含量为98.58%,栽培宁前胡总挥发油含量为93.48%;万家乡栽培宁前胡总挥发油含量为99.50%;宁国市栽培宁前胡(统货)总挥发油含量为95.89%。宁前胡挥发油中共鉴定出78个成分,烯烃类为主要化学成分,不同生长条件下宁前胡中烯烃类含量分别为野生宁前胡65.26%,中溪镇栽培宁前胡89.88%,万家乡栽培宁前胡62.08%,栽培宁前胡(统货)66.11%;所有烯烃类成分中,α-蒎烯的含量均最高。结论野生和栽培宁前胡中挥发油成分差异较大,可以通过闻"气"辨别野生前胡与栽培前胡。Objective To investigate the content of volatile oil components in Ningguo Peucedanum praeruptorurn Dunn under different growth conditions using gas chromatography-mass spectrometry (GC- MS) and the differences in the content and type of volatile oil in wild and cultivated Ningguo Peucedanum praeruptorum Dunn. Methods The wild and cultivated Peucedanurn praeruptorum Dunn in Zhongxi Town of Ningguo, cultivated Peucedanurn praeruptorum Dunn in Wanjiaxiang Town of Ningguo, and the gradeless and uniformly-priced goods of cultivated Peucedanum praeruptorum Dunn in Ningguo were used as samples. Volatile oil was extracted using steam distillation in The Pharmacopoeia of The People's Republic of China Volume I. GC-MS was used to analyze the chemical components of volatile oil, and chromatographic peak area normalization was used to calculate the relative content of each component. Results The total content of volatile oil was 98. 58% in wild Peucedanum praeruptorurn Dunn and 93.48% in the cultivated Peucedanum praeruptorum Dunn in Zhongxi Town of Ningguo, 99.50% in the cultivated Peucedanum praeruptorurn Dunn in Wanjiaxiang Town of Ningguo, and 95.89% in the gradeless and uniformly-priced goods of cultivated Peucedanum praeruptorum Dunn in Ningguo. A total of 78 components were identified in the volatile oil of Ningguo Peucedanum praeruptorum Dunn, and the main chemical components were olefins. The content of olefins in Ningguo Peucedanum praeruptorum Dunn under different growth conditions was 65.26% in wild Peucedanum praeruptorum Dunn, 89.88% in cultivated Peucedahum praeruptorum Dunn from Zhongxi Town, 62.08% in Peucedanum praeruptorum Dunn from Wanjiaxiang Town, and 66. 11% in cultivated Peucedanum praeruptorum Dunn (gradeless and uniformlypriced goods). Of all olefins, alpha-pinene had the highest content. Conclusion There are significant differences in the content of volatile oil components between wild and cultivated Ningguo Peucedanum praeruptorum Dunn, and wild and cultivated Peucedanum praeruptorum Du
