摘  要:目的探索不同麻醉状态对Wistar大鼠图形视觉诱发电位(pattern visual evoked potential,PVEP)稳定性的影响。方法 40只8~10周龄Wistar大鼠,麻醉后于初级视皮层对应位置硬脑膜表面埋置记录电极,于鼻骨处埋置参考电极。对麻醉状态下的大鼠采用时间频率为2 Hz和空间频率为0.04 c·d^(-1)的黑白翻转棋盘格刺激,记录相同时间段内的PVEP及基线脑电(electroencephalogram,EEG)。根据EEG功率频谱分析将麻醉分为多个阶段,比较不同阶段大鼠PVEP振幅及峰值期的稳定性。结果麻醉状态下大鼠的EEG可以根据功率频谱图分为delta波为主的慢波睡眠期(0.1~3.0 Hz)、过渡期(3.1~4.4Hz)、theta波为主的浅睡眠期(4.5~6.0 Hz)。三个阶段的PVEP振幅分别为(20.55±8.32)μV、(32.25±4.86)μV、(28.12±4.79)μV,振幅的变异系数分别为40.49%、15.08%、17.04%。结论不同的麻醉状态下记录到的PVEP的振幅及峰值期稳定性不同。记录麻醉动物的PVEP时,应当同时监测实验动物的EEG活动情况,并且建议在EEG活动表现为浅麻醉状态时进行。Objective To explore the effects of different anesthetized states on the stability of pattern visual evoked potential (PVEP) in Wistar rats. Methods Forty 8 to 10 weeks old Wistar rats were used in this experiment. Firstly,the recording elec- trodes were put on the dura surface corresponding to the primary visual cortex after anesthesia. In the meantime, the reference electrodes were buried in the nasal bone of rats. Then black and white checkerboard with time frequency of 2 Hz and spatial frequency of 0.04 c · d-1 were used to stimulate the anesthetized rats. At last the PVEP and baseline electroencephalogram (EEG) during the same time period were recorded. The anesthetized state was divided into several stages by the analysis of EEG power spectral. The stability of PVEP in different stages were compared. Results The anesthetized state of rats could be divided into three stages according to the EEG power spectrum: slow wave sleep ( delta wave ,0.1 - 3.0 Hz), the transition period ( 3.1 - 4.4 Hz), light sleep ( theta wave, 4.5 - 5.0 Hz ). PVEP amplitudes during the three stages were (20.55 ±8.32) μV, (32.25±4.86) μN, (28.12 ±4.79) μV, respectively. The varia- tion coefficients of amplitudes were 40.49% , 15.08% and 17.04% , respectively. Conclusion The stability of PVEP recorded under different anesthetized states are different. When recording PVEP in anesthetized animals, EEG activity of animals should be monitored simultaneously, and the best state for recording is under the light anesthesia.
