摘 要:安康串状核桃为2014年陕西审定通过的省级林木良种,结实成串状,平均每串结果6.1个,品质较好,丰产性强,了解安康串状核桃的亲缘关系及推测其起源,对于指导核桃育种有重要意义。利用SSR技术对串状核桃亲缘关系分析研究,以核桃幼嫩叶片为材料,用文献所得多态性SSR引物对同为核桃属的10种材料的基因组DNA进行扩增,并进行聚类分析。结果表明,10对引物在供试材料上共扩增出96条带,每对引物扩增出的DNA条带数为3~16条。遗传分析表明,10种核桃属材料间的遗传距离为0.027 4~0.814 3,在10种材料中,串状核桃和西洛3号的遗传距离最小,为0.257 5,说明串状核桃与西洛3号的亲缘关系最近。'Ankangchuan' a cultivar of Juglans regia was examined and approved at provincial level in Shaanxi in 2014. The cultivar demonstrated many excellent cultivation features, such as bunched fruit (with an average number of fruit Of 6.1) ,high yield,and high fruit quality. It is of importance to understand genetic relationship, and to predict its biological origin to guide the breeding of walnut. Tender leaves of the walnut cultivars were used as materials. Genomic DNA of 10 kinds of walnut materials was amplified by the polymorphic SSR primers which were from the references. Clustering analysis was conducted. Ninty six different bands were amplified from 10 pairs of high polymorphic primers. The number of the bands from each pair of primer was 3 to 16. The genetic analysis showed that the genetic distances between the 10 walnut materials were 0. 027 4-0. 814 3. Among the 10 kinds of walnut materials, 'Ankangchuan' and 'Xiluo 3' had the lowest genetic distance of 0. 257 5,indicating that these two cultivars had the closest genetic relationship.
